摘 要:为研究不同盐度对哈维弧菌生理生化和耐药性的影响,探讨不同盐度对哈维弧菌的耐药表型与携带耐药基因的变化,以哈维弧菌9H7R为主要研究对象,在盐度为5、10、15、20、25 ppt下,采用纸片扩散法(Kirby-Bauer纸片扩散法,简称K-B法),参照NCCLS抗生素敏感试验操作标准,研究哈维弧菌在不同盐度下耐药性的变化。然后提取细菌RNA、经反转录及qPCR,再根据特定基因计算相对表达量。结果显示:不同盐度生长的哈维弧菌,对喹诺酮类药物的耐药性发生了变化,且耐药基因gyrB的表达量受到了影响。For the study of different salinity effect on physiological, biochemical and drug resistance of V.harveyi, we investigatec the changes of drug-resistant phenotype and drug-resistant gene of V.harveyi at different salinity using V.harveyi 9H7R;the salinity of seawater was 5, 10, 15, 20,25 ppt, and Kirby-Bauer paper diffusion method was performed. According to NCCLS antibiotic sensitivity test operation standard, the change of drug resistance of V.harveyi in different salinity was studied, then bacterial RNA was extracted, reversed transcription and qPCR were performed,and relative expression levels were calculated according to specific genes. The results showed that Vibrio harveyi grew at different salinities.The resistance to quinolones changed and the expression of gyr B was affected.
