Salt Tolerance Identification and Molecular Marker Evaluation of 57 Wheat Micro-core Germplasms at Seedling Stage

摘  要:为了进一步发掘小麦耐盐种质并将其应用于分子育种,为其高效利用提供分子辅助选择依据,以耐盐品种旱选10号与盐敏感品种中国春作为对照,通过苗期耐盐指数(Salt injury index,SI)对57份小麦微核心种质进行表型鉴定,同时利用已报道的24个耐盐性状相关分子标记对其进行标记。结果表明,在250 mmol/L NaCl溶液胁迫后,供试种质的盐害指数呈正态分布,表现为高感、敏感、中耐、耐盐、高耐的种质分别有5、21、24、6、1份,其中,评价等级为耐盐及以上(SI<40.00%)的种质有带芒红麦、黄瓜先、半截芒、小偃6号、兴义4号、Atlas 66和尚麦等7个品种,且带芒红麦、黄瓜先和半截芒的耐盐性明显高于对照旱选10号(SI=30.00%),尤其是带芒红麦为高耐种质(SI=14.24%);Xcfd9在供试种质中具有a、b、c等3种标记型,对应的种质平均盐害指数分别为49.59%(中耐)、57.06%(中耐)和64.74%(盐敏感),存在显著差异(F>F_(0.05),P=0.04),表明Xcfd9与供试种质耐盐性存在紧密相关,且a型或b型标记型在研究筛选出的7份耐盐种质中均存在。To further explore the wheat salt-tolerant germplasm and apply it to molecular breeding,provide the basis for molecular assisted selection for the efficient utilization of molecular breeding,in this study,phenotypic identification of 57 wheat micro-core germplasms was conducted by the salt injury index(SI)using salt-tolerant variety Hanxuan 10 and salt-sensitive variety Chinese Spring as the controls.Simultaneously,the tested germplasms were marked with 24 reported salt-tolerance-related molecular markers.The results showed that SI of the tested germplasms displayed a normal distribution after stress by 250 mmol/L of NaCl solution,and the number of germplasms showing high sensitive,sensitive,medium tolerant,tolerant,and high tolerant were 5,21,24,6,and 1,respectively.Among the germplasms,7 varieties with excellent salt tolerance(SI<40.00%)were Daimanghongmai,Huangguaxian,Banjiemang,Xiaoyan 6,Xingyi 4,Atlas 66,and Heshangmai.The salt tolerance grade of Daimanghongmai,Huangguaxian,and Banjiemang was obviously higher than that of the control Hanxuan 10(SI=30.00%),especially Daimanghongmai was a high-tolerant germplasm(SI=14.24%).Marker analysis results showed that Xcfd9 had 3 genotypes a,b,and c among the tested germplasm,and their corresponding average SI was 49.59%(medium tolerant),57.06%(medium tolerant),and 64.74%(sensitive),respectively,showing a significant difference,which indicated that Xcfd9 was closely related to salt tolerance of the tested germplasms,and the a-or b-genotype existed in all of the 7 salt-tolerant germplasms screened in this study.
