摘  要:病程相关基因1的非表达子(NPR1)是植物抗病信号传导的关键基因,在植物响应病原菌侵染过程中发挥重要作用。为分析谷子NPR1家族基因的特征及响应禾生指梗霉侵染的表达水平,根据已报道的拟南芥、水稻NPR1蛋白序列,利用BLASTP对xiaomi全基因组进行鉴定,利用生物信息学方法进行理化性质分析、进化分析、基因结构分析,利用转录组数据分析谷子NPR1家族基因的组织表达特异性及响应禾生指梗霉胁迫下的表达特征。结果表明,共筛选到谷子NPR1家族基因4个,分别位于第5、8、9号染色体上,依次命名为SiNPR1~SiNPR4,编码475~629个氨基酸,其中,SiNPR1定位于细胞核中,SiNPR2、SiNPR4定位于叶绿体中,SiNPR3定位于质膜且为唯一的疏水蛋白。进化分析发现,谷子与拟南芥、水稻NPR1家族基因可分为3类,其中,SiNPR2和SiNPR4聚为一支,SiNPRs与OsNPRs亲缘关系更近。motif分析中共鉴定到8个motif基序,除SiNPR3仅含有5个保守基序外,其余成员均含8个保守基序,且排列顺序均为motif6、motif2、motif7、motif3、motif4、motif5、motif1、motif8;顺式作用元件分析发现,该家族成员主要含有水杨酸、茉莉酸甲酯、赤霉素等激素元件。组织特异性分析发现,仅SiNPR1在不同时期未成熟种子中特异高表达,其余基因均在不同时期叶片中高表达;转录组分析发现,SiNPR1在受禾生指梗霉侵染的谷子叶片和穗中均上调表达,表明SiNPR1可能在抵御禾生指梗霉侵染方面发挥重要作用。Non-expressor of pathogenesis-related gene 1(NPR1)is a key gene involved in disease-resistant signal transduction of plants,which plays an important role in the process of plant response to pathogen infection.To analyze the characteristics of foxtail millet NPR1 family genes(SiNPRs)and their expression levels in response to Sclerospora graminicola infection,in this study,SiNPRs were identified in the whole genome of xiaomi aligning according to the reported protein sequences in Arabidopsis thaliana(AtNPRs)and in Oryza sativa(OsNPRs)via BLASTP.The physicochemical properties,phylogenetic relationship,and gene structure of SiNPRs were analyzed using bioinformatics tools.The tissue expression specificity and expression characteristics in response to S.graminicola stress of SiNPRs were analyzed based on the transcriptome data.The results showed that four SiNPRs were identified,which were located on chromosomes 5,8 and 9,respectively.They were named SiNPR1-SiNPR4,encoding 475-629 amino acids.Among them,SiNPR1 was located in the nucleus,SiNPR2 and SiNPR4 were located in the chloroplast.SiNPR3 was located in the plasma membrane and was the only hydrophobic protein.Phylogenetic analysis showed that the SiNPRs were divided into three categories with Arabidopsis thaliana and Oryza sativa.SiNPR2 and SiNPR4 were clustered into one,and SiNPRs was more closely related to OsNPRs.A total of 8 motifs were identified by motif analysis.Except that SiNPR3 contained only 5 conserved motifs,other SiNPRs genes involved 8 conserved motifs,which were motif6,motif2,motif7,motif3,motif4,motif5,motif1,motif8 in turn.Cis-acting element analysis showed that SiNPRs mainly contained salicylic acid(SA),methyl jasmonate(MJ),gibberellin(GA)and so on.Tissue-specific analysis showed that only SiNPR1 was highly expressed in immature seeds at different stages,and the remaining genes were highly expressed in leaves at different stages.Transcriptome analysis showed that SiNPR1 was up-regulated in leaves and spikes of foxtail millet infected by Sclerospor
