摘 要:为进一步提高甘薯植株再生及脱毒效率,获得适宜甘薯的高效脱毒技术,为甘薯健康种薯种苗的培育及产量、品质提升奠定基础,以兼用型甘薯品种郑红23号为试验材料,以其茎尖分生组织为外植体,培养基设置7个处理,即MS(空白对照)、MS+0.1 mg/L NAA+1~6 mg/L 6-BA培养获得再生植株,依次对SPLCV、SPCSV、SPFMV、SPLV和SPVG等5种主要病毒病进行PCR检测,通过比较各处理的植株再生率和脱毒率,分析2种激素不同浓度配比对郑红23号茎尖植株再生及脱毒效率的影响。结果表明,在含有NAA和6-BA激素处理下,郑红23号茎尖分生组织再生率均高于90%,且极显著高于空白对照,其中在MS+0.1 mg/L NAA+4 mg/L 6-BA培养基上再生率最高,为97.78%,此时再生植株5种病毒脱毒率最高,为13.60%。在MS+0.1 mg/L NAA+1 mg/L 6-BA上培养的再生植株生长速度最快,且随着6-BA浓度的升高,再生苗生长速度逐渐变慢。值得注意的是,MS+0.1 mg/L NAA+6 mg/L 6-BA的愈伤诱导率高达100%,该处理下甘薯再生率和脱毒率分别为91.11%和11.79%,但整体而言,郑红23号茎尖再生的脱毒率较低。To further improve the efficiency of plant regeneration and detoxification of sweet potato,obtain a highly efficient detoxification technology for sweet potato,and lay the foundation for cultivation of healthy seed potato and seedlings,and improvement of yield and quality of sweet potato,in this study,Zhenghong 23,a variety of sweet potato both edible and processed,was used as the experimental material,and its shoot apical meristem was used as the explant.The regenerated plants were obtained by using 7 treatments of culture mediums including MS(control)and MS+0.1mg/L of NAA+1-6 mg/L of 6-BA.Five major virus diseases including SPLCV,SPCSV,SPFMV,SPLV,and SPVG were detected by PCR successively.The regeneration rate and virus-free rate of the plants of treatments were compared to analyze the effects of different concentration ratio of the two hormones on shoot apical plant regeneration and virus-free efficiency of Zhenghong 23.The results indicated that the regeneration rate from shoot apical meristem of Zhenghong 23 was higher than 90%under the treatments of NAA and 6-BA,which was significantly higher than that of control.The medium of MS+0.1 mg/L of NAA+4 mg/L of 6-BA was the optimal medium on which regeneration rate was the highest,and it was 97.78%.In this case,the virus-free rate of the five viruses in the regenerated plants was the highest,and it was 13.60%.In addition,the fastest growth was achieved under the medium of MS+0.1 mg/L of NAA+1 mg/L of 6-BA.The growth rate of regenerated seedlings gradually slowed down with the increase of 6-BA concentration.It was worth noting that the callus induction rate was up to 100%cultured on the medium of MS+0.1 mg/L of NAA+6 mg/L of 6-BA,and the regeneration rate and virus-free rate of sweet potato under this treatment were 91.11%and 11.79%,respectively.In general,the virus-free rate of shoot apical regeneration of Zhenghong 23 was low.
