摘  要:目的探讨缺氧缺血性损伤对新生猪脑内钠钾ATP酶及CaMKⅡ表达的影响。方法28头雄性新生约克夏猪随机分为对照组(进行假手术,n=4)、模型组(建立缺氧缺血脑损伤模型,n=24)。模型组根据缺氧缺血时间均分成6个亚组(0~2 h组、2~6 h组、6~12 h组、12~24 h组、24~48 h组、48~72 h组),每组4头。利用免疫荧光染色法对各组脑切片进行染色,比较各组钠钾ATP酶、CaMKⅡ的表达状况。结果各组均见钠钾ATP酶阳性表达,6~12 h组表达最低;相邻两两比较结果显示,0~2 h组与2~6 h组、2~6 h组与6~12 h组、6~12 h组与12~24 h组、12~24 h组与24~48 h组钠钾ATP酶表达均有统计学差异(均P<0.05)。钠钾ATP酶表达呈先下降后上升然后继续下降的趋势。各组均见CaMKⅡ阳性表达,6~12 h组表达最低;0~2 h组与2~6 h组、2~6 h组与6~12 h组、6~12 h组与12~24 h组、24~48 h组与48~72 h组CaMKⅡ表达有统计学差异(均P<0.05),CaMKⅡ表达呈现先减低然后增高,然后再减低再增高改变。结论随着时间的推移,缺氧缺血后新生猪钠钾ATP酶表达呈“横S型”趋势,而CaMKⅡ表达呈“双谷双峰”样改变。Objective To investigate the effects of hypoxic ischemic on the expression of Na+,K+-ATPase and CaMKⅡin the brains of newborn piglets.Methods Twenty-eight newborn male Yorkshire pigs were randomly divided into control(sham operation only,n=4)and experimental groups(hypoxic ischemic injury model,n=24).The experimental group was divided into 6 subgroups according to the time after ischemic hypoxic injury(0-2 h group;2-6 h group;6-12 h group;12-24 h group;24-48 h group;48-72 h group),4 in each group.The expression of Na+,K+-ATPase and CaMKⅡwere compared by immunofluorescence staining of brain slices from each group.Results Positive expression of Na+,K+-ATPase was seen in all groups,and reached its lowest point in the 6-12 h group.There were significant differences between groups 0-2 h and 2-6 h,2-6 h and 6-12 h,6-12 h and 12-24 h,and 12-24 h and 24-48 h(all P<0.05).The expression of Na+,K+-ATPase decreased first,then increased,and later continued to decrease.Positive expression of CaMKⅡwas found in all groups,and the lowest point was seen in the 6-12 h group,and there were significant differences between groups 0-2 h and 2-6 h,2-6 h and 6-12 h,6-12 h and 12-24 h,and 24-48 h and 48-72 h(all P<0.05).The expression of Na+,K+-ATPase decreased first,then increased and afterwards continued to decrease.Conclusion The expression of Na+,K+-ATPase and CaMKⅡchanged with time following hypoxic ischemic injury.The expression of Na+,K+-ATPase after hypoxic ischemic injury fit the shape of“horizon S type”,and the expression of CaMKⅡafter hypoxic ischemic injury fit the pattern of“double valley and double peaks”.
