摘 要:本研究利用生物信息学方法对高粱GATA基因家族成员进行全基因组鉴定和分析,以期为其基因功能研究奠定基础。结果表明,在高粱基因组中共鉴定到30个GATA基因,不均匀地分布在8条染色体上,2号和7号染色体无分布;26个基因含有CX_(2)CX_(18)CX_(2)C保守基序,SbGATA3、SbGATA4、SbGATA18和SbGATA30含有CX_(2)CX_(20)CX_(2)C保守基序;进化树分析表明,高粱GATA基因家族可分为4组,分别包含15、9、4、2个基因,同一组内的成员保守锌指结构域蛋白序列一致性更高,基因结构和保守基序高度相似;高粱GATA蛋白与单子叶植物玉米和谷子的同源性大于双子叶植物拟南芥;顺式调控元件分析发现,SbGATA启动子序列中存在调控生长发育、激素信号传导和逆境胁迫响应元件,检测到光响应元件G-box、脱落酸响应元件ABRE、茉莉酸甲酯响应元件CGTCA-motif和厌氧诱导响应元件ARE数量最多,另外检测到低温响应元件LTR和干旱诱导性元件MBS。表达分析表明,SbGATA16、SbGATA9、SbGATA25和SbGATA20在多数组织中表达量均较高。模拟干旱处理高粱GATA基因转录组数据分析发现,SbGATA11和SbGATA26上调表达,说明这两个基因可能在干旱胁迫响应中发挥重要作用。The bioinformatics method was used to identify the GATA family genes in the whole genome of sorghum,in order to lay foundations for research on functions of SbGATA genes.Total of 30 SbGATA members were identified,which distributed unevenly on 8 chromosomes,and none was on chromosome 2 and 7.Among which,26 SbGATA genes contained the CX_(2)CX_(18)CX_(2)C conserved domain,while SbGATA3,SbGATA4,SbGATA18 and SbGATA30 contained the CX_(2)CX_(20)CX_(2)C conserved domain.The analysis results of evolutionary tree showed that they could be divided into 4 groups including 15,9,4 and 2 genes respectively,and the type,order and motifs were similar in proteins within the same group.The homology of sorghum GATA proteins to monocot Zea mays and Setaria italica was greater than Arabidopsis.The analysis of cis-elements showed that the SbGATA promoter sequence contained the elements which regulating growth and development,hormone signaling and stress response.The number of light responsive element G-box,ABA-responsive element ABRE,methyl jasmonate responsive element CGTCA-motif and anaerobic induction responsive element ARE were more than others.In addition,low temperature response element LTR and drought-inducibility element MBS were found.Expression analysis showed that the expression level of SbGATA16,SbGATA9,SbGATA25 and SbGATA20 were higher in most tissues.The results of drought stress under PEG6000 showed that the expression of SbGATA11 and SbGATA26 were up-regulated under drought stress,speculating they had important roles in response to drought stress.
