摘 要:为了调查猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)在河南省的流行情况及变异规律,试验对2018—2019年河南省39家猪场送检的175份猪肺脏、脾脏、肝脏和淋巴结等样本进行PRRSV的PCR检测,对阳性样本扩增ORF5和NSP2基因,并进行遗传进化分析、氨基酸序列比对,同时预测ORF5基因的N-糖基化位点。结果表明:PRRSV的阳性检出率为25.71%(45/175),从45份阳性样本中扩增得到16个ORF5和10个NSP2基因序列。基于ORF5基因推导氨基酸序列的系统发育分析,检出的PRRSV阳性样本中有16株属于基因2型,可进一步划分为4个谱系;这16株PRRSV毒株在跨膜区(TM)、主要中和表位(PNE)、诱饵表位和N-糖基化位点上存在差异,免疫受体酪氨酸抑制基序(ITIM)在77~82 aa区域高度保守。基于NSP2基因推导氨基酸序列的系统发育分析,检出的PRRSV阳性样本中有10株均属于基因2型,可进一步划分为2个谱系,并且其中类NADC30 PRRSV毒株在322~432 aa、483 aa和504~522 aa处有不连续的氨基酸缺失。说明2018—2019年河南地区PRRSV优势毒株是类NADC30 PRRSV毒株,且ORF5和NSP2基因发生较大变异。In order to investigate the prevalence and variation of Porcine reproductive and respiratory syndrome virus(PRRSV)in Henan Province,the experiment conducted PRRSV PCR detection on 175 pig lung,spleen,liver and lymph node samples from 39 farms in Henan Province from 2018 to 2019.The ORF5 and NSP2 genes were amplified from positive samples,and genetic evolution analysis,amino acid sequence comparison,and prediction of the N-glycosylation site of ORF5 gene was performed.The results showed that the positive detection rate of PRRSV was 25.71%(45/175),and 16 ORF5 and 10 NSP2 gene sequences were amplified from 45 positive samples.Based on the phylogenetic analysis of the deduced amino acids of the ORF5 gene,16 of the detected PRRSV positive samples belonged to genotype 2,which could be further divided into 4 lineages.These 16 PRRSV strains differed in transmembrane region(TM),predominant neutralizing epitope(PNE),decoy epitope and N-glycosylation sites(NGSs);immunoreceptor tyrosine inhibitory motif(ITIM)was highly conserved in region 77-82 aa.Based on the phylogenetic analysis of the deduced amino acids of the NSP2 gene,10 of the detected PRRSV positive samples belonged to genotype 2,which could be further divided into 2 lineages.Among them,the NADC30-like PRRSV strains had discontinuous amino acid deletions at 322-432 aa,483 aa and 504-522 aa.The results suggested that the dominant PRRSV strain in Henan Province from 2018 to 2019 was the NADC30-like PRRSV strain,and the ORF5 and NSP2 genes had large mutations.
