摘 要:目的从2013—2020年麻疹核酸阳性标本中分离获得单纯疱疹1型病毒(herpes simplex virus type1,HSV-1),并分析其基因特征。方法将2439份麻疹核酸阳性咽拭子标本接种到Vero/SLAM细胞上进行病毒分离培养,出现细胞病变后收获,提取核酸,并经实时荧光定量-聚合酶链式反应(qPCR)、PCR及测序进行分析。结果共获得30株HSV-1,经系统发育分析,30株HSV-1序列gG区的核苷酸和氨基酸同源性分别为95.58%~100%和90.57%~100%,均属于A基因型。结论HSV-1在四川省广泛分布且可能来源于同一传播链。麻疹病毒感染增加了HSV-1的感染风险,或是麻疹病毒感染激活了潜伏的HSV-1感染还需进一步研究。Objective To analyze the genetic characteristics of herpes simplex virus type 1(HSV-1)isolated from measles positive cases from 2013 to 2020.Methods 2439 throat swab samples tested positive for measles nucleic acid were inoculated into Vero/SLAM cells for virus isolation.The isolates were collected and nucleic acid was extracted,and analyzed by quantitative real-time polymerase chain reaction(qPCR),PCR and sequencing.Results A total of 30 HSV-1 strains were obtained.Sequences from gG gene of HSV-1 were used to classify the isolates into genotype A.The homology of nucleotide and amino acid was 95.58%-100%and 90.57%-100%,respectively.Conclusion HSV-1 is widely distributed in Sichuan province,and the prevalence of HSV-1 may have originated from the same transmission chain.Whether measles virus infection increases the risk of HSV-1infection or activates latent HSV-1 infection needs further studying.
