摘 要:目的:研究水杨酰芳胺衍生物N-(3-氯-4-氟苯基)-2-羟基-4-(3-(哌啶-1-基)丙氧基)苯甲酰胺(NPBM)和N-(3-氯-4-氟苯基)-2-羟基-4-(3-吗啉丙氧基)苯甲酰胺(NMBM)通过作用于STAT-3对肝癌HepG-2细胞的影响。方法:采用不同浓度化合物NPBM、NMBM作用于肝癌HepG-2细胞,氯硝柳胺为阳性对照药,MTT比色法检测细胞存活率。采用不同浓度的化合物NPBM和NMBM作用于HepG-2肝癌细胞,荧光免疫试验检测对HepG-2肝癌细胞中STAT-3表达水平的影响。结果:不同浓度NPBM和NMBM处理后,肝癌HepG-2细胞的存活率逐渐下降,化合物氯硝柳胺、NPBM和NMBM对肝癌HepG-2细胞的log C分别为-4.627,-4.092,-4.314。结论:化合物NPBM和NMBM能够抑制肝癌细胞,作用机制可能与抑制STAT-3信号通路有关。Objective:This paper studies the effects of NPBM and NMBM on hepatoma HepG-2 cells by acting on Stat-3.Method:Different concentrations of NPBM and NMBM were used to act on hepatoma HepG-2 cells.Niclosamide was used as the positive control drug.The cell survival rate was detected by MTT colorimetry.Different concentrations of NPBM and NMBM were used to act on HepG-2 hepatoma cells.The expression level of STAT-3 in HepG-2 hepatoma cells was detected by fluorescence immunoassay.Result:After treatment with different concentrations of NPBM and NMBM,the survival rate of hepatoma HepG-2 cells decreased gradually.The log C of compounds niclosamide,NPBM and NMBM on hepatoma HepG-2 cells were-4.627,-4.092 and-4.314,respectively.Conclusion:Compounds NPBM and NMBM can inhibit hepatoma cells,and the mechanism may be related to the inhibition of STAT-3 signaling pathway.
