摘  要:核酸回收是生物化学实验课程中一项基础的实验技术,然而目前实验教学中普遍采取商品化试剂盒开展实验,且有关RNA的回收实验鲜有涉及。因此,枣庄学院生物化学教研室设计新型核酸回收实验,可直接用于DNA的回收;装置经去RNA酶处理后也可对RNA片段进行回收。该实验的具体操作为:首先制备融胶装置,利用离心机的高速离心力结合陶瓷纤维棉,实现核酸凝胶切割融化;然后通过酚仿抽提蛋白,去除杂质;最后用无水乙醇沉淀核酸。本实验装置制作简单,回收过程简便,回收产物可用于核酸的分子克隆等研究。该实验可增强教学内容设计的启发性和互动性,不仅可加强学生实践和创新能力、培养学生科研思维,也可以为生物化学实验教学中开设综合设计实验奠定基础。The recovery of DNA or RNA fragment from agarose gel is a one of the rudimentary and most commonly used in in molecule biological study.At the present time,the common use method to recover nucleic acid is agarose gel DNA extraction kit.In addition,there are currently no special technology for the recovery of RNA.Therefore,this project designs the economical,rapid and efficient method for reclaiming DNA or RNA from agarose gel.Firstly,the agarose gel was melted by centrifuge combine with ceramic fiber bulk.And then removes impurities using chloroform.Finally,the nucleic acid is precipitated with anhydrous alcohol.By this means,only through high speed centrifugation and ethanol precipitate can obtain the DNA or RNA fragment.This experiment put students together in an inspiring and creative intellectual environment where they are encouraged to pursue research.On the other hand,it can promote the cultivation of innovative talents,and lay the foundation for the comprehensively innovative experiment in biochemistry experimental teaching.
