摘  要:目的基于网络药理学探讨真武汤治疗慢性心力衰竭和慢性肾功能不全的“异病同治”的作用机制。方法根据口服生物利用度(OB)和类药性(DL)通过中药系统药理学平台(TCMSP)筛选真武汤的有效成分及靶基因,并整合通过Pubchem数据库及Swiss数据库获取的靶基因,采用基因组注释数据库平台(GeneCards)预测慢性心力衰竭及慢性肾功能不全作用靶点,并运用Uniprot数据库查询相关基因名称。将药物与疾病交集靶点利用String和Cytoscape进行蛋白互作(protein-protein interaction,PPI)网络构建及关键共有靶点筛选,并进行GO富集分析和KEGG富集分析。结果本研究共筛选得符合标准活性成分61个,药物潜在作用靶点569个,慢性心力衰竭疾病靶点1751个,慢性肾功能不全靶点1395个,取交集后获得药物-疾病共有靶点150个,通过Cytoscape拓扑分析得到核心靶点54个,富集于生物过程、细胞组成、分子功能及112条信号通路上,通路主要涉及炎性反应、血管内皮再生、血流动力学、细胞凋亡等,这些通路可能是真武汤治疗慢性心力衰竭和慢性肾功能不全的活性途径。结论研究预测出真武汤治疗慢性心力衰竭及慢性肾功能不全“异病同治”的机制主要涉及炎性反应、血管内皮再生、血流动力学、细胞凋亡等相关信号通道,为进一步实验研究和临床运用提供新的思路及参考依据。Objective Based on network pharmacology to explore the"same treatment for different diseases"mechanism of Zhenwu Decoction(真武汤)in the treatment of chronic heart failure and chronic renal insufficiency.Methods According to the oral bioavailability(OB)and drug-like properties(DL),the active ingredients and target genes of Zhenwu Decoction were screened through the Traditional Chinese Medicine System Pharmacology Platform(TCMSP),and the target genes obtained through the Pubchem database and the Swiss database were integrated,and the genome annotation database was used.The platform(GeneCards)predicted the targets of chronic heart failure and chronic renal insufficiency,and used the Uniprot database to query related gene names.String and Cytoscape were used to construct the protein-protein interaction(PPI)network,screen the key shared targets for the intersection targets of drugs and diseases,and perform GO enrichment analysis and KEGG enrichment analysis.Results In this study,a total of 61 active ingredients that met the standard,569 potential drug targets,1751 chronic heart failure disease targets,1395 chronic renal insufficiency targets were screened,and 150 drug-disease targets were obtained after the intersection.54 core targets were obtained through Cytoscape topology analysis,which were enriched in biological processes,cell composition,molecular functions and 112 signaling pathways,which mainly involved inflammatory response,vascular endothelial regeneration,hemodynamics,apoptosis,etc.These pathways may be the active pathways of Zhenwu Decoction in the treatment of chronic heart failure and chronic renal insufficiency.Conclusion This study predicted that the"same treatment for different diseases"mechanism of Zhenwu Decoction in the treatment of chronic heart failure and chronic renal insufficiency mainly involves related signaling channels such as inflammatory response,vascular endothelial regeneration,hemodynamics and apoptosis.The application provides new ideas and references.
