摘 要:为探寻可能引发鸡支气管阻塞的病毒类病原,试验采用RT-PCR方法检测了自河北及周边地区收集的43份有支气管阻塞病料样品中H5亚型禽流感病毒(AIV)、H7亚型AIV、H9亚型AIV、传染性支气管炎病毒(IBV)和新城疫病毒(NDV)等病毒类病原感染情况。结果显示,阳性样品共24份,总阳性率为55.8%(24/43),其中H9亚型AIV、IBV、H5亚型AIV、H7亚型AIV、NDV的阳性率分别为34.9%(15/43)、20.9%(9/43)、0(0/43)、0(0/43)、0(0/43),不存在H9亚型AIV和IBV的混合感染;自检测阳性样品中分离、鉴定了5株H9N2 AIV,HA蛋白裂解位点模式均为PSRSSR↓GLF,属于低致病性毒株,位于h9.4.2.5分支,3株分离毒株HA蛋白关键氨基酸位点模式与疫苗毒株WJ57完全一致,但5株分离毒株NA蛋白关键氨基酸位点模式与疫苗毒株WJ57不完全一致;自检测阳性样品分离、鉴定了4株IBV,S1蛋白裂解位点模式包括HRHRR和HRRRR,属于LX4型,均与疫苗毒株YX10 D90亲缘关系较近,与疫苗毒株Connecticut和H120亲缘关系较远。结果提示,部分地区发生支气管阻塞鸡群存在H9亚型AIV或IBV的单独感染,2种病毒分离毒株与致病性、抗原性密切相关的基因均存在有别于疫苗毒的分子特征。In order to explore the viral pathogens that may cause bronchial obstruction in chickens, H5 subtype avian influenza virus(AIV), H7 subtype AIV, H9 subtype AIV, infectious bronchitis virus(IBV) and Newcastle disease virus(NDV) were detected by RT-PCR in 43 samples collected from chicken flocks with bronchial obstruction in Hebei province and neighboring region. The results showed that 24 positive samples were detected, the total positive rate was 55.8%(24/43). The positive rate of H9 subtype AIV, IBV, H5 subtype AIV, H7 subtype AIV, and NDV was 34.9%(15/43), 20.9%(9/43), 0(0/43), 0(0/43), and 0(0/43),respectively. Mixed infection of H9 subtype AIV with IBV was not found. The HA protein cleavage site pattern of 5 H9 subtype AIV strains identified from positive samples was PSRSSR↓GLF, and HA protein genes were belonged to the h9.4.2.5 branch.The key amino acid sites pattern of HA protein of 3 isolated strains was completely consistent with that of vaccine strain WJ57.While the key amino acid sites pattern of NA protein of 5 isolated strains was not completely consistent with that of vaccine strain WJ57. The S1 protein cleavage site patterns of 4 IBV strains identified from positive samples included HRHRR and HRRRR.The results of genetic evolution analysis showed that the 4 IBV strains were belonged to LX4 type and closely related to vaccine strain YX10 D90, and distantly related to vaccine strain Connecticut and H120. The results indicated that single infection of H9 subtype AIV or IBV circulated in diseased chickens with bronchial obstruction, and molecular and antigenic characteristics to distinguish the two type isolates from vaccine viruses was demonstrated.
