摘 要:为复合制备沙门菌SP4噬菌体微囊化微球并评估其治疗效果,本研究采用液滴法以明胶/海藻酸钠/壳聚糖为原料制备SP4噬菌体微囊化微球,并测定SP4噬菌体微囊化微球的效价、酸碱稳定性、在模拟胃液中的稳定性、不同温度下的保存稳定性、在模拟肠液中释放行为等生物学特性,以及SP4噬菌体微囊化微球治疗雏鸡沙门菌感染的效果。结果显示,当明胶浓度为8 g/100 mL、海藻酸钠浓度为2 g/100 mL、壳聚糖浓度为0.4 g/100 mL时,SP4噬菌体微囊化微球的效价最佳;SP4噬菌体微囊化微球在pH5~pH8范围内非常稳定;SP4噬菌体微囊化微球在模拟胃液中能保持较好的活性;SP4噬菌体微囊化微球在4℃保存时具有较好的稳定性;SP4噬菌体微囊化微球在模拟肠液中能够较好的释放。雏鸡沙门菌感染治疗试验结果显示,SP4噬菌体微囊化微球比SP4噬菌体悬浊液治疗效果好,感染组雏鸡经SP4噬菌体微囊化微球治疗后存活率为90%,而未治疗感染组雏鸡全部死亡。与未治疗感染组相比,治疗感染组雏鸡脏器未发生明显病理变化。上述结果表明SP4噬菌体微囊化微球有严格的宿主特异性,裂解能力强,能够显著提高感染沙门菌雏鸡的存活率。本研究首次制备的SP4噬菌体微囊化微球能够裂解致病性沙门菌,为噬菌体的临床抗菌应用提供参考依据。In order to explore the compound preparation and therapeutic effect of Salmonella SP4 phage, SP4 phage microencapsulated microspheres were prepared by drop method with gelatin/sodium alginate/chitosan as raw materials. The biological characteristics of SP4 phage microencapsulated microspheres, such as titers, acid-base stability, simulated gastric juice stability, preservation stability and simulated intestinal juice release behavior, were determined. And the effect of SP4 phage microencapsulated microspheres on Salmonella infection in chicks was evaluated. The results showed that the titer of SP4 phage microencapsulated microspheres was the best when the concentrations of gelatin, sodium alginate, and chitosan were 8g/100mL,2g/100mL and 0.4g/100mL, respectively. SP4 phage microencapsulated microspheres were very stable in the range of pH5-p H8.SP4 phage microencapsulated microspheres can preserve good activity in simulated gastric juice. SP4 phage microencapsulated microspheres had good stability when stored at 4℃. SP4 phage microencapsulated microspheres can be well released in simulated intestinal juice. The results of treatment of Salmonella-infected chicks with SP4 phages showed that SP4 phage microencapsulated microspheres had better therapeutic effect than SP4 phage suspension. The survival rate of infected chicks treated with SP4 phage microencapsulated microspheres was 90%, while all the infected groups died. Compared with the infection group, the treatment group did not cause significant pathological changes in chicken organs. The results showed that SP4 phage microencapsulated microspheres had strict host specificity and strong lysis ability, which could significantly improve the survival of chicks infected with Salmonella. The SP4 phage microencapsulated microspheres prepared in this study can lyse pathogenic Salmonella, which provides a reference basis for promoting the clinical antibacterial application of phage.
