金果胃康胶囊对PLGC大鼠Wnt/Lgr5信号通路的干预研究

摘　　要：目的探讨金果胃康胶囊对胃癌前病变大鼠细胞信号传导通路中Wnt1、β-catenin、Lgr5基因及其蛋白表达的干预机制。方法选用90只雄性SD大鼠,随机分为空白组12只,造模组78只,采用MNNG结合浓盐水灌胃及饥饱失常等综合造模方法复制PLGC模型,确定造模成功后随机分为模型组、中药高、中、低剂量治疗组及维酶素治疗组,分别给予生理盐水、不同剂量的金果胃康胶囊、维酶素,治疗6w。采用HE染色、AB-PAS染色光镜下观察大鼠胃黏膜病理改变情况;RT-PCR法检测Wnt/Lgr5信号通路中Wnt1、β-catenin、Lgr5基因及蛋白表达情况。结果与模型组相比,金果胃康胶囊治疗组大鼠胃黏膜上皮组织病理改变情况有所改善,Wnt1 mRNA、β-cateninmRNA、Lgr5mRNA表达量均显著下降,有统计学意义(P<0.01)。结论金果胃康胶囊可通过降低大鼠胃黏膜的Wnt1、β-catenin、Lgr5的蛋白表达及基因转录,促进细胞凋亡,降低胃癌发生的可能,达到治疗PLGC的目的。Objective To explore the effect of Jinguo Weikang Capsule on intervention mechanism of Wnt1,β-catenin,Lgr5 genes and their protein expression.Methods 90 male SD rats were randomly divided into blank group(12 rats)and model group(78 rats).The PLGC model was reproduced by MNNG combined with concentrated saline gavage and abnormal hunger and satiety.After the model was established successfully,they were randomly divided into model group,high,medium and low dose treatment groups of traditional Chinese medicine and vitamin treatment group.They were given normal saline,different doses of Jinguo Weikang capsule and vitamin for 6 weeks.HE staining and AB-PAS staining were used to observe the pathological changes of rat gastric mucosa under light microscope;Detection of Wnt1,β-catenin,Lgr5 gene and protein expression in Wnt/Lgr5 signal pathway by RT-PCR.Results Compared with the model group,the pathological changes of gastric mucosal epithelium in the Jinguo Weikang Capsule treatment group were improved,and the expression of Wnt1 mRNA,β-catenin mRNA,Lgr5 mRNA decreased significantly(P<0.01).Conclusion Jinguo Weikang capsule can reduce the protein expression and gene transcription of Wnt1,β-catenin and Lgr5,promote cell apoptosis and reduce the possibility of gastric cancer,so as to achieve the purpose of treating PLGC.
