摘 要:为延长药物在体内的循环时间,基于超支化聚(酰胺-胺)独特的超支化结构,结合索拉非尼对肿瘤生长明显的抑制效果,构建了索拉非尼/超支化聚(酰胺-胺)的载药体系。为评估载药体系的缓释效果,利用高效液相色谱法对索拉非尼的体外释放过程进行在线检测,从而获得其动力学参数,并推算出药物缓释的拟合方程。结果证明,索拉非尼在该载药体系中前12 h为快速释放期,整个药物释放完全需要约50 h。In order to improve the circulation time of drugs in the body,a sorafenib/hyperbranched poly(amide-amine)drug delivery system is constructed based on the unique structure of the hyperbranched polymer and the sorafenib’s obvious inhibition on tumor.The high performance liquid chromatography(HPLC)method is applied to evaluate the sustained-release effect for the drug-loading system.The released drug is detected and quantified at different time to obtain the kinetic parameters,and the drug releasing equation.It is proved that sorafinib is released in vitro fast in the first 12 hours,and the complete release of the drug will take about 50 hours.
