摘　　要：目的:开展远志-石菖蒲药对不同提取物对PC12细胞保护作用研究,探索远志和石菖蒲联用的用量配比。方法:采用谷氨酸损伤PC12细胞建立模型,MTT法测定细胞活力、Hoechst 33342染色液/PI双染法检测细胞凋亡和细胞坏死情况。结果:远志-石菖蒲合用效果较佳,水提和醇提物均能明显提高细胞活力并抑制细胞凋亡。结论:远志-石菖蒲干粉按1∶4的比例合用对谷氨酸致神经细胞损伤的保护作用较强。Objective:To study the effects of different extracts of Polygala tenuifolia and Acorus tatarinowii on PC12 cell apoptosis,and to explore the dosage ratio of Polygala tenuifolia and Acorus tatarinowii.Methods:Using glutamate to build a cell injury model,the cell viability is measured by MTT method,and the apoptosis and necrosis are detected by Hoechst 33342 staining solution/PI double staining.Results:The combination of Polygala tenuifolia and Acorus tatarinowii had better effect.Both water extract and alcohol extract could significantly improve cell viability and inhibit cell apoptosis.Conclusion:The combination of Polygala tenuifolia and Acorus tatarinowii in the ratio of 1∶4 has a strong protective effect on neuronal apoptosis induced by glutamate.
