摘　　要：目的:提供一种应用Sartobind Q膜去除乙脑灭活疫苗中细胞残留DNA的方法。方法:使用传统的Sepharose 6 Fast Flow层析所获得的初步纯化液进行Sartobind Q膜层析,在一定的离子强度和pH下,使DNA吸附在Sartobind Q膜的配基上,而病毒蛋白不吸附直接从膜下流出,得到最终纯化液,通过抗原含量和宿主细胞残留DNA的含量评价纯化结果。结果:用Sartobind Q膜处理后疫苗残留DNA比初步纯化液减少95.4%~97.8%,而且病毒抗原含量无显著变化。结论:Sartobind Q膜可吸附乙脑疫苗中Vero细胞DNA残留,进一步降低疫苗杂质含量。Objective:The objective is to provide a method for removing residual DNA in cells of JE inactivated vaccine with Sartobind Q membrane.Methods:The Sartobind Q membrane chromatography is performed on the preliminary purified liquid obtained by traditional Sepharose 6 Fast Flow chromatography.At a certain ionic strength and pH value,the DNA is adsorbed on the artobind Q membrane ligand,and the viral protein is not adsorbed directly from the membrane outflow,and the final purification solution is obtained.Purification results are evaluated by antigen content and host cell residual DNA content.Results:After treatment with Sartobind Q membrane,the residual DNA of the vaccine are reduced by 95.4%~97.8%compared with the preliminary purified solution,and the content of virus antigen are not significantly changed.Conclusion:Sartobind Q membrane has the ability to adsorb Vero cell DNA residues in JE vaccine and further reduce vaccine impurities content.
