摘 要:目的探讨金丝桃苷(Hyp)联合核因子-κB(NF-κB)抑制剂对肺炎小鼠肺指数及sB7-H3水平的影响。方法60只4~6周龄SPF级BALB/c小鼠按照随机数字表法分为4组,每组各15只,选取其中45只采用脂多糖口咽吸入法建立肺炎模型,Hyp干预组进行50 mg/kg Hyp灌胃和0.9%氯化钠溶液皮下注射;联合干预组进行50 mg/kg Hyp灌胃和5μmol/LNF-κB抑制剂BAY11-7082皮下注射;对照组和肺炎模型组进行等剂量0.9%氯化钠溶液灌胃和皮下注射,均连续干预8周。分别于干预2、5和8周,称重并计算小鼠肺指数;干预8周,采用酶联免疫吸附试验(ELISA)试剂盒检测4组BALF和肺组织匀浆sB7-H3、肿瘤坏死因子α(TNF-α)和白细胞介素-8(IL-8)水平,蛋白质印迹法检测肺组织Akt和NF-κB蛋白磷酸化水平,并计算p-Akt/Akt和p-NF-κB/NF-κB比值。结果HYP干预组和联合干预组肺指数均随干预时间显著降低(P<0.05)。干预2周,与对照组相比,肺炎模型组、Hyp干预组和联合干预组小鼠肺指数均显著增加,差异均有统计学意义(P<0.05),而后3组差异无统计学意义(P>0.05);而干预5周,联合干预组肺指数较肺炎模型组显著增加,差异有统计学意义(P<0.05),但2个治疗组相比差异无统计学意义(P>0.05);干预8周后,4组间两两相比差异均有统计学意义,且联合干预组肺指数也显著低于HYP干预组(P<0.05),即将该时间点作为后续指标检测最佳时间点。干预8周,与对照组相比,肺炎模型组、Hyp干预组和联合干预组BALF及肺组织sB7-H3、TNF-α和IL-8水平、肺组织Akt和NF-κB蛋白磷酸化水平均显著增加,差异均有统计学意义(P<0.05);与肺炎模型组相比,Hyp干预组和联合干预组BALF及肺组织sB7-H3、TNF-α和IL-8水平、肺组织Akt和NF-κB蛋白磷酸化水平均显著降低,且联合干预组以上指标也较Hyp组显著降低,差异均有统计学意义(P<0.05)。结论经金丝桃苷联合NF-κB抑制剂联合干预后,肺炎小鼠肺指数、BALF和肺组�Objective To investigate the effect of hypericin(Hyp)combined with nuclear factor-κB(NF-κB)inhibitor on lung index and sB7-H3 level in mice with pneumonia.Methods Sixty SPF grade BALB/c mice aged 4-6 weeks were divided into 4 groups according to the random number table method,15 mice in each group.45 of them were selected to establish a pneumonia model by LPS oropharyngeal inhalation method,and the Hyp intervention group was given 50 mg/kg Hyp by gavage and subcutaneous injection of normal saline;the combined intervention group received 50 mg/kg Hyp by gavage and 5μmol/L NF-κB inhibitor BAY11-7082 subcutaneously;the control group and the pneumonia model group were given the same dose of normal saline by intragastric gavage and subcutaneous injection,and both were continuously intervened for 8 weeks.At 2 weeks,5 weeks and 8 weeks of the intervention,the mice were weighed and the lung index was calculated;after 8 weeks of intervention,the levels of sB7-H3,tumor necrosis factor alpha(TNF-α)and interleukin-8(IL-8)in BALF and lung tissue homogenates in 4 groups were detected by enzyme-linked immunosorbent assay(ELISA)kits.The phosphorylation levels of Akt and NF-κB proteins in lung tissue were detected by Western blotting,and the ratios of p-Akt/Akt and p-NF-κB/NF-κB were calculated.Results The lung index in both the HYP intervention group and the combined intervention group decreased significantly with the intervention time(P<0.05).After 2 weeks of intervention,compared with the control group,the lung index of the mice in the pneumonia model group,the Hyp intervention group and the combined intervention group were significantly increased,the differences were statistically significant(P<0.05),while the difference in the latter three groups was not statistically significant(P>0.05).However,after 5 weeks of intervention,the lung index in the combined intervention group was significantly higher than that in the pneumonia model group,the difference was statistically significant(P<0.05),but there was no significant
