摘 要:目的:观察右美托咪定对老年小鼠剖腹探查术所致神经炎症的影响。方法:根据随机数字表,将48只老年C57BL/6小鼠(16月龄)随机分成假手术组、手术组、右美托咪定组、右美托咪定+手术组,每组12只。右美托咪定组腹腔注射25μg/kg右美托咪定;手术组行剖腹探查术;美托咪定+手术组在行剖腹探查术前20分钟预先腹腔注射25μg/kg右美托咪定。假手术组腹腔注射等体积0.9%生理盐水。剖腹探查术后8小时,使用ELISA试剂盒检测各组小鼠海马组织白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)水平;Western blot检测各组小鼠海马组织IL-1β、IL-6、TNF-α表达情况。结果:与假手术组比较,手术组小鼠海马组织炎症因子(IL-1β、IL-6、TNF-α)明显表达升高(P<0.05),而与手术组相比,右美托咪定+手术组老年小鼠海马组织促炎细胞因子(IL-1、IL-6、TNF-α)明显表达降低(P<0.05)。假手术组与右美托咪定组海马组织促炎细胞因子(IL-1β、IL-6、TNF-α)无统计学差异(P>0.05)。结论:右美托咪定能改善老年小鼠剖腹探查术所致神经炎症。Objective:To explore the effect of dexmedetomidine on exploratory laparotomy-induced neuroinflammation in aged mice.Methods:According to the table of random digit,forty-eight C57BL/6 mice(16-month-old)were randomly divided into four groups:Sham group,Dexmedetomidine group,Surgery group and Dexmedetomidine+Surgery group,with 12 mice in each group. Mice in Dexmedetomidine group received intraperitoneal injection with 25 μg/kg dexmedetomidine. Mice in Surgery group received the procedure of exploratory laparotomy. Mice in Dexmedetomidine+Surgery group was intraperitoneally injected 25 μg/kg dexmedetomidine 20 minutes before conducting exploratory laparotomy. Sham group received intraperitoneal injection ofequal volume 0.9% normal saline without conducting exploratory laparotomy. Eight hours after finishing exploratory laparotomy,ELISA test kit was employed to measure the expression level of IL-1β,IL-6,TNF-αin hippocampi. Moreover,the expression level of pro-inflammatory cytokine(IL-1β,IL-6,TNF-α)in hippocampi was measured by Western blot.Results:(1)Compared with Sham group,pro-inflammatory cytokine(IL-1β,IL-6,TNF-α)in the hippocampi of Surgery group was significantly increased(P<0.05). When compared with the Surgery group,pro-inflammatory cytokine(IL-1β,IL-6,TNF-α)in the hippocampi ofDexmedetomidine+Surgery groupwas significantly decreased(P<0.05). No significant difference was observed in the expression of pro-inflammatory cytokine in hippocampi between Sham group and Dexmedetomidine group(P>0.05).Conclusion:Dexmedetomidine can ameliorate exploratory laparotomy-induced neuroinflammation in aged mice.
