摘  要:目的:通过检测咪喹莫特诱导的银屑病小鼠模型皮损VEGFR-1及VEGFR-2的表达水平,为研究银屑病血管机制方向提供实验基础。方法:Balb/c小鼠随机分为实验组(n=7)和正常对照组(n=5),实验组使用咪喹莫特乳膏诱导建立银屑病小鼠模型。免疫组化染色检测VEGFR-1及VEGFR-2在小鼠皮肤中的表达及分布,并对VEGFR-1和VEGFR-2染色阳性的血管进行计数以及与血管、表皮厚度之间的相关性。结果:实验组表皮、真皮血管与毛囊均过度表达VEGFR-1(P<0.05)和VEGFR-2(P<0.05);实验组VEGFR-1血管染色阳性率较VEGFR-2血管染色阳性率高(P<0.05);实验组VEGFR-1及VEGFR-2的免疫组化染色累积光密度值(IOD值)分别与小鼠微血管数量、表皮厚度存在正相关(P<0.05)。结论:银屑病小鼠皮损中VEGFR-1与VEGFR-2过度表达,其表达强度与总血管数量、表皮角质形成细胞的增殖程度呈正相关。Objective:To evaluate the influence of VEGFR-1 and VEGFR-2 in the imiquimod-induced mouse model.Methods:The Balb/c mice were divided into the experimental(n=7)and the control groups(n=5),respectively. In the experimental group,imiquimod cream was used to induce psoriatic mouse model,while the control group was used vaseline cream. Cutaneous VEGFR-1 and VEGFR-2 protein expressions were assessed with immunohistochemical staining. The positive stained vessels among the two groups were counted. And the correlation testsbetween the staining densities and the vessel numbers as well as epidermalthicknesswerewere analyzed.Results:The positively stained cells of VEGFR-1(P<0.05)and VEGFR-2(P<0.05)were mostly distributed in the epidermis,the upper dermis and the follicles. The correlation test showed that the density of VEGFR-1 and VEGRF-2 expression were positively correlated with that of vessels(P<0.05)and epidermal thickness(P<0.05).Conclusion:Imiquimod-induced psoriatic mouse model express higher levers of both VEGFR-1 and VEGFR-2 in the epidermis,the upper dermis and the follicles,which are positively correlated with vessel density and the proliferation of the epidermal keratinocytes respectively.
