摘  要:氟苯尼考(florfenicol,FFC)是目前临床上应用广泛的动物广谱抗菌药,本研究旨在采用乳化溶剂挥发法制备氟苯尼考纳米晶(florfenicol nanocrystal,FFC-NC)。通过单因素试验优化制备工艺,即以FFC纳米混悬液的粒径大小作为筛选的主要参数,对6个影响因素进行考察,确定最佳制备条件,并对在最优条件下制备的FFC-NC进行物理表征评价。最终确定FFC-NC制备的最佳条件:有机相与水相体积比为1∶2,泊洛沙姆188的浓度为5‰,匀浆速度和时间分别为7000 r/min和5 min,匀质压力和次数分别为300 bar和3次。利用最佳条件制备得到的FFC-NC平均粒径为(226.1±11.3)nm,PDI平均值为0.29±0.03。FFC-NC的粒径分布在141.8~243.0 nm范围内,表明其粒径分布范围窄。FFC-NC的理化性质表征结果显示,其化学结构未发生改变,纳米晶形态为不规则球体,粒径小且分布均一,纳米晶的晶型结构发生改变。电位测定结果表明,FFC纳米混悬液的稳定性好。本研究突破了传统的FFC-NC制备技术,为其在兽医临床上的使用提供了一种新的方法。Florfenicol(FFC)is a broad-spectrum animal antibacterial drug widely used clinically.The purpose of this study is to prepare florfenicol nanocrystals(FFC-NC)using the emulsion solvent evaporation method.By single factor test analysis,the hydrodynamic size of FFC-NC was used as the main parameter for screening the optimal preparation conditions.Physicochemical characterization of FFC-NC prepared under the optimal preparation condition were evaluated.The results showed that the best condition for the preparation of FFC-NC was as follows:volume ratio of organic phase and aqueous phase of 1∶2,concentration of P188 of 5‰,speed of homogenization at 7000 r/min,time of homogenization of 5 min,pressure of homogenization of 300 bar,and number of homogenization cycles as 3.The average particle size of FFC-NC prepared under the best condition was(226.1±11.3)nm and the average polymer dispersity index(PDI)was 0.29±0.03.The particle size of most particles ranges 141-243 nm,and the particle size distribution was narrow.Results of physicochemical characterization showed that the chemical structure of FFC-NC had not changed,the morphology of FFC-NC was an irregular sphere,and the particle size distribution was uniform and the crystal structure of FFC-NC was changed.Results of potential measurement showed that the suspension of florfenicol nanocrystals had good stability.In conclusion,this study broke through the traditional preparation technology of FFC-NC and provided a new method for the clinical use of FFC by veterinarians.
