摘 要:胰淀素(amylin)兼具有发挥抑食与促温双重作用。中枢能量调控系统中,可卡因苯丙胺调节转录肽(cocaine-and amphetamine-regulated transcript,CART)、神经型一氧化氮合成酶(neuronal nitric oxide synthase,nNOS)因其具诱导厌食和维持体质量成为治疗肥胖潜在靶点。目前鲜有证据表明Amylin能量调控与CART、nNOS神经元有关,为补充这一数据,本试验探寻CART和nNOS脑内定位,以及Amylin对于CART、nNOS神经元表达数目的调控作用,该效应发生是否具依赖交感神经系统(aympathetic nervous system,SNS)。结果显示,CART分布于下丘脑背内侧核(dorsomedial hypothalamic nucleus,DMH),动眼神经副核(pre-edinger-westphal nucleus,PrEW)和前包钦格复合体(pre-botzinger complex,PrBO)等核团,而nNOS在背侧背盖核(lateral dorsal tegmental nucleus,LDT)有明显阳性神经元胞体,且在DMH存在共定位。Amylin注射显著促进下丘脑CART(P=0.0183)和nNOS(P=0.0308)表达,提示CART与nNOS涉及Amylin的调节过程,并依赖于完整的背肩胛棕色脂肪(interscapular brown adipose tissue,IBAT)SNS。通过研究Amylin与中枢食欲相关神经元间的关系,为探究能量稳态与产热调控中枢作用关系提供积极证据。Amylin has the dual effects of suppressing food and promoting temperature.In the central energy regulation system,Cocaine-and amphetamine-regulated transcript(CART)and neuronal Nitric oxide synthase(nNOS)have become potential treatments for obesity due to their ability to induce anorexia and maintain body weight.Target.At present,there is little evidence that Amylin?s energy regulation is related to CART and nNOS neurons.To supplement this data,this experiment explored the localization of CART and nNOS in the brain.Amylin?s regulatory effect on the number of expressions of CART and nNOS neurons.Whether the effect is dependent on the Sympathetic nervous system(SNS).The results showed that CART is distributed in Dorsomedial hypothalamic nucleus(DMH),Pre-Edinger-Westphal nucleus(PrEW)and Pre-Botzinger complex(PrBO),etc.Nucleus,and nNOS has obvious positive neuron cell bodies in the Laterodorsal tegmentalnucleus(LDT)region,and co-localization in DMH.Amylin injection significantly promotes hypothalamic CART(P=0.0183),nNOS(P=0.0308)expression,suggesting that CART and nNOS are involved in the regulation of Amylin,and this effect depends on the intact Interscapular brown adipose tissue(IBAT)SNS.In this paper,by studying the relationship between Amylin and central appetite-related neurons,it provides some evidence for exploring the mechanism of the central role of energy homeostasis and thermogenesis regulation.
