摘 要:2020-2021年在河南省18个地区共收集499份发生腹泻的仔猪临床样品,采用RT-PCR进行猪流行性腹泻病毒(porcine epidemic diarrhea virus,PEDV)筛查。结果显示,共检测出162份阳性样品,阳性率为35.2%。对其中21株PEDV流行毒株S基因进行了测序、同源性比对和遗传进化分析,结果显示21株PEDV流行毒株之间核苷酸同源性为96.9%~100%,氨基酸同源性为95.2%~100%。与52株2011-2019年国内流行毒株之间的核苷酸同源性为95.1%~99.5%,氨基酸同源性为91%~99.8%。遗传进化结果显示,21株PEDV毒株均属于GⅡb群。氨基酸序列比对显示,流行毒株S蛋白存在7处新的且规律的氨基酸突变。本试验对现阶段我国所流行的PEDV S基因进行测序分析,为了解PEDV流行毒株遗传变异趋势和研发新的有效疫苗提供参考。In order to monitor the prevalence and variation of porcine epidemic diarrhea virus(PEDV)in Henan Province,a total of 499 clinical samples of piglets with diarrhea were collected from 2020 to 2021 in Henan Province.The PEDV screening was performed by RT-PCR.The results showed that a total of 162 positive samples were detected,with a positive rate of 35.2%.The S gene in 21 PEDV strains were sequenced,then the homologous alignment and genetic evolution analysis were performed.The results of homology alignment showed the nucleotide homology were 96.9%-100%and amino acid homology were 95.2%-100%among 21 PEDV strains.From 2011 to 2019,the nucleotide homology and amino acid homology of 52 domestic circulating strains were 95.1%-99.5%and 91%-99.8%respectively.The results of genetic evolution showed that all the 21 PEDV strains belonged to GⅡb group.Amino acid sequence alignment showed that there were 7 new and regular amino acid mutations in the S protein of the epidemic strain.In this study,the PEDV S genes prevailing in China at present were sequenced and analyzed,which can provide reference for understanding the genetic variation trend of PEDV epidemic strains and developing new effective vaccines.
