摘 要:目的考察半枝莲提取物对HepG2肝癌细胞的抑制作用及作用机制。方法体外培养HepG2肝癌细胞,MTT法考察半枝莲提取物对HepG2肝癌细胞增殖的影响;TRANSWELL法考察察半枝莲提取物对HepG2肝癌细胞侵袭和迁移的影响;透射电镜观察半枝莲提取物对HepG2肝癌细胞自噬的影响;Western-Blot法考察半枝莲提取物对HepG2肝癌细胞Raf/MEK/ERK信号通路蛋白的影响。结果0、1.6、3.2、6.25、25、50、100μg/mL和200μg/mL半枝莲提取物孵育后,HepG2肝癌细胞生存率分别为(100.3±3.2)%、(97.8±11.9)%、(88.2±11.1)%、(65.3±8.7)%、(40.9±5.8)%、(27.3±4.0)%、(17.3±1.0)%、(8.3±0.5)%;半枝莲提取物可诱发HepG2肝癌细胞的自噬;20μg/mL半枝莲提取物组迁移率显著高于0μg/mL半枝莲提取物组(t=10.255,P=0.000);20μg/mL半枝莲提取物组侵袭率显著高于0μm半枝莲提取物组(t=8.260,P=0.001);半枝莲提取物可剂量依赖性地抑制p-MEK和p-ERK的表达。结论半枝莲提取物可抑制HepG2肝癌细胞的增殖、侵袭和迁移,诱发细胞的自噬,该作用与半枝莲提取物抑制Raf/MEK/ERK信号通路有关。Objective To investigate the inhibitory effect and mechanism of Banzhilian(Scutellaria barbata)extract on HepG2 hepatoma cells.Methods HepG2 hepatoma cells were cultured in vitro,and the effects of Banzhilian(Scutellaria barbata)extract on proliferation of HepG2 hepatoma cells were investigated by MTT method.The invasion and migration of HepG2 hepatoma cells were observed by Transwell method.The autophagy of HepG2 hepatoma cells was observed by transmission electron microscope.The Raf/MEK/ERK signal transduction of Banzhilian(Scutellaria barbata)extract on HepG2 hepatoma cells was detected by Western blot.Results The survival rates of HepG2 hepatoma cells were(100.3±3.2)%,(97.8±11.9)%,(88.2±11.1)%,(65.3±8.7)%,(40.9±5.8)%,(27.3±4.0)%,(17.3±1.0)%and(8.3±0.5)%respectively after incubated with 0μg/mL,1.6μg/mL,3.2μg/mL,6.25μg/mL,25μg/mL,50μg/mL,100μg/mL and 200μg/mL Banzhilian(Scutellaria barbata)extract,respectively.The extract of Banzhilian(Scutellaria barbata)could induce autophagy in HepG2 cells.The migration rate of 20μg/mL Banzhilian(Scutellaria barbata)extract group was significantly higher than that of 0μg/mL Banzhilian(Scutellaria barbata)group(t=10.255,P=0.000),and the invasion rate of 20μg/mL Banzhilian(Scutellaria barbata)extract group was significantly higher than that of 0μg/mL Banzhilian(Scutellaria barbata)extract group(t=8.260,P=0.001).Banzhilian(Scutellaria barbata)extract could inhibit the expressions of p-mek and p-ERK in a dose-dependent manner.Conclusion Banzhilian(Scutellaria barbata)extract can inhibit the proliferation,invasion and migration of HepG2 hepatoma cells and induce autophagy,which is related to the inhibition of Raf/MEK/ERK signaling pathway.
