摘 要:鸭坦布苏病毒是一种黄病毒科黄病毒属单股正链RNA病毒,可造成蛋鸭、种鸭产蛋下降,给养鸭业造成巨大经济损失的重要疾病。目前,坦布苏弱毒活疫苗是临床上有效预防该病的重要手段之一。CpG ODN作为疫苗佐剂被证实能够有效提高疫苗的免疫效果,但其对弱毒活疫苗的是否具有免疫增强的效果尚不清楚。本研究选择鸭坦布苏弱毒活疫苗FX2010-180P,以CpG ODN为免疫佐剂,在鸭体内进行免疫以及攻毒保护实验。通过检测鸭体内抗体水平、免疫保护率、组织切片来进行免疫效果的评估。结果显示,FX2010-180P免疫组鸭子在免疫后3~4 d即可产生特异性抗体,而添加CpG ODN疫苗佐剂组,免疫鸭的抗体产生时间滞后于未添加组。攻毒保护实验结果显示,两种方式免疫方式都可以保护鸭子不受坦布苏病毒野毒的感染。本研究结果显示添加CpG ODN对鸭坦布苏FX2010-180P弱毒活疫苗并不具有免疫增强的效果。Duck Tambusu virus(DTMUV)belongs to the family Flavivirus and is a positive-sense single-stranded RNA virus,which causes severe egg production drop in egg-laying ducks and huge economic losses to duck industry.Live attenuated DTMUV vaccine is one of the important methods to effectively prevent the disease.CpG ODN,as a vaccine adjuvant,has been proved to be effective in enhancing the immune effect of vaccines.However,whether CpG ODN enhances the immune effect of live attenuated vaccine is unknown.In this study,the enhancement effect of CpG ODN on the attenuated live DTMUV vaccine(FX2010-180P)was evaluated by examining the antibody level and protection in vaccinated ducks.The results showed that ducks immunized with FX2010-180P alone induced specifi c antibodies on 3-4 days post immunization while ducks immunized with FX2010-180P combined with CpG ODN adjuvant showed delayed se roconversion.Both of vaccines with and without CpG ODN adjuvant protected the vaccinated duck against challenge with the virulent DTMUV.All data showed that CpG ODN did not enhance the immune response of DTMUV attenuated live vaccine in ducks.
