摘 要:以实验室保存的奶牛乳房炎源金黄色葡萄球菌为宿主菌,从新疆石河子地区奶牛场粪便和污水中分离纯化裂解性噬菌体,并对其生物学特性进行分析,为奶牛乳房炎的噬菌体防治提供材料,并通过噬菌斑、透射电子显微镜观察,测定其最佳感染复数、噬菌谱、热稳定性、pH稳定性、生长曲线等特性进行分析。结果显示:分离到1株裂解性噬菌体P42,噬菌斑呈透亮无晕环的圆形,透射电镜观察形态为短尾噬菌体;目前此噬菌体只能裂解其宿主菌,最佳感染复数为0.001;60℃20 min时活性丧失,且在pH4.0~9.0时,噬菌体P42的活性没有明显的变化;感染宿主菌的潜伏期是10 min,暴发期为140 min,裂解量约为126 PFU/cell。本研究分离筛选到的噬菌体P42是一株裂解性短尾科噬菌体,其裂解效果较好,繁殖迅速,为奶牛乳房炎的噬菌体防治及其制剂的研发提供了参考。In order to provide materials for the phage control of bovine mastitis,Staphylococcus aureus from a mastitis case kept in our laboratory was used as host bacteria for isolation and characterization of lytical bacteriophage from feces and sewages on a dairy farm in Shihezi area,Xinjiang.The morphology of the isolated bacteriophage was observed by plaque and transmission electron microscope,and its other characteristics were also examined including optimal multiplicity of infection,host range,thermal stability,pH stability,one-step growth curve, etc. The results showed that the isolated lytic bacteriophage P42 strain formed circular plaques with bright rings but without halo and appeared as short-tailed phage in morphology under transmission electron microscope. At present, this bacteriophage only lysed the host S. aureus . The optimal multiplicity of infection was 0.001. Activity was lost at 60℃ for 20 min, and it was no signifi cant change at pH4.0-9.0. The incubation period of the infected host bacteria was 10 min, the outbreak period was 140 min, and the burst size was about 126 PFU/cell. In conclusion, the bacteriophage p42 isolated in this study was a lytical Podoviridae with good lysis effect and rapid propagation. It provided a reference for the phage control of mastitis and the development of its preparation.
