摘  要:目的探讨银杏叶提取物(EGB)对乳腺癌MCF-7细胞增殖、焦亡的影响及作用机制。方法将质量浓度为0、10、20、40、80、160 mg/L的EGB作用于体外培养的人乳腺癌MCF-7细胞上,观察细胞形态;四甲基偶氮唑蓝染色法(MTT)检测细胞增殖;Annexin V/PI双染色流式细胞术检测细胞焦亡率;乳酸脱氢酶(LDH)释放实验检测LDH释放量;Western Blot检测MCF-7细胞中Calpain-1、NLRP3、Th17、Treg、caspase-1、cleaved caspase-1、GSDMD的表达。结果EGB可明显抑制MCF-7细胞的增殖;增加MCF-7细胞的焦亡率;增加MCF-7细胞的LDH释放量;增加GSDMD、caspase-1、cleaved caspase-1蛋白表达;降低Calpain-1、NLRP3、Th17、Treg蛋白表达;且作用随着EGB浓度的增加逐渐增强。结论银杏叶可抑制乳腺癌细胞的增殖和迁移,其可能与Calpain-1/NLRP3调控的细胞焦亡及Th17/Treg炎症轴相关。Objective To investigate the effects and mechanism of extract of Ginkgo biloba(EGB)on the proliferation and apoptosis of breast cancer MCF-7 cells.Methods EGB(concentration of 0,10,20,40,80 and 160 mg/L)were applied to human breast cancer MCF-7 cells cultured in vitro.Cell proliferation was assessed by tetramethylazolium blue staining(MTT).Cell pyroptosis was assessed by Annexin V/PI double staining flow cytometry.lactate dehydrogenase(LDH)release was assayed by LDH kit.The protein expression of Calpain-1,NLRP3,Th17,Treg,caspase-1,cleaved caspase-1 and GSDMD in MCF-7 cells were assess by Western Blot.Results EGB could significantly inhibit the proliferation of MCF-7 cells.It also increased the pyroptosis and LDH release in MCF-7 cells.In addition,EGB increased the expression of GSDMD,caspase-1 and cleaved caspase-1 protein while down-regualted the expression of Calpain-1,NLRP3,Th17 and Treg proteins.The effect was gradually enhanced with the increase of EGB concentration.Conclusion Ginkgo biloba can inhibit the proliferation of breast cancer cells,which may be related to Calpain-1/NLRP3 regulation of cell pyrosis and Th17/Treg inflammatory axis.
