摘  要:【目的】明确山羊痘病毒(GTPV)AV41弱毒疫苗株在不同日龄山羊原代睾丸细胞中的增殖效率,为选择病毒或疫苗株的培养细胞提供数据支撑。【方法】通过传统方式对7日龄和60日龄山羊睾丸细胞分离培养,利用长时间动态细胞成像及数据分析系统比较2个日龄细胞的生长效率;接种GTPV,观察2个日龄细胞在感染病毒后的形态特征;通过间接免疫荧光和荧光定量PCR比较GTPV的增殖效率。【结果】7日龄和60日龄山羊睾丸细胞的形态特征和生长速率无明显差异;接种GTPV后,2个日龄的原代山羊睾丸细胞均能产生细胞病变,但是出现细胞病变的时间以及病变特征存在差别。间接免疫荧光和荧光定量PCR结果表明:GPTV在60日龄山羊睾丸细胞中的增殖效率高于7日龄。【结论】GTPV在7日龄和60日龄山羊原代睾丸细胞中均能发生病毒增殖,且2种细胞均能发生明显的细胞病变,但60日龄细胞的增殖效率高于7日龄细胞。研究结果对采用山羊睾丸细胞培养GTPV弱毒株有一定的指导作用。[Purpose]To determine the proliferation efficiency of goat pox virus(GTPV)AV41 attenuated vaccine strain in primary testicular cells of goats at different ages,and to provide datas to support the selection of cultured cells for virus or vaccine strains.[Methods]Two ages of cells(7-day-old and 60-day-old goat testicular cells)were isolated and cultured in traditional way.The growth efficiency of the two ages of cells was compared by the long-term dynamic cell imaging and data analysis system,and then GTPV was inoculated to observe the morphological characteristics.The propagation efficiency of GTPV was compared by indirect immunofluorescence(IFA)and fluorescent quantitative PCR(qPCR).[Results]There was no obvious differences in morphological characteristics and growth rate between the two ages of cells.After GTPV inoculation,the two ages of primary goat testicular cells could produce cytopathic effect(CPE),but there were differences in the occurrence time and CPE characteristics.IFA and qPCR results showed that the propagation effi-ciency of GTPV in 60-day-old goat testicular cells was higher than that in 7-day-old goat testicular cells.[Conclusion]Both 7-day-old and 60-day-old goat primary testicular cells can develop obvi-ous CPE after inoculation with GTPV.GTPV can proliferate in both cells,but the propagation effi-ciency is different.These results may be useful for the culture of GTPV attenuated strain with goat testis cells.
