摘 要:目的探讨经头皮下应用氯化锰(MnCl_(2))溶液在MRI神经成像中的可行性,并确定最佳给药浓度。方法在头皮下颅骨表面滴涂不同浓度的MnCl_(2)溶液构建模型,选取18只SD大鼠随机分为3组(A、B、C),各组分别使用浓度为100、250、500 mM的MnCl_(2)溶液,用于研究锰离子增强磁共振成像(ME-MRI)中Mn^(2+)溶液的最佳显像浓度。另选取12只大鼠作为加钙组,将CaCl_(2)加入到MnCl_(2)溶液中,随机分为2组(D组使用400 mM CaCl_(2)+100 mM MnCl_(2),E组使用250 mM CaCl_(2)+250 mM MnCl_(2)),以研究离子强度对输送效率的可能影响。于滴涂后3 h和24 h进行MRI检查。结果经颅施加Mn^(2+)的渗透性越接近颅缝位置的Mn^(2+)渗透越好。高浓度的MnCl_(2)(500 mM)会产生较大的对比噪声比(CNR)(P<0.001),但24 h后能清晰显示海马、丘脑等区域;低浓度(100 mM)的溶液则不能跨越颅骨。在添加Ca^(2+)后,低浓度的MnCl_(2)溶液能透过颅骨,且其传输效率与相应的Ca^(2+)浓度成正比(P<0.001)。经颅应用500 mM浓度的MnCl_(2)后在24 h时进行ME-MRI对神经组织的显示效果最好。结论一定浓度的Mn^(2+)溶液可以通过完整的颅骨进入脑组织内,且添加Ca^(2+)后能有效提高Mn^(2+)的穿透能力。经头皮下给药的ME-MRI对神经组织有较好的显示效果,此发现可为动物模型中神经通路的Mn^(2+)成像研究提供一种侵袭性相对较小的方法,开拓了Mn^(2+)在神经成像中的应用。Objective To explore the feasibility and the optimal dose of MnCl_(2) solution in MRI neurography by transcranial method.Methods 18 SD rats were enrolled and randomly divided into three groups(A,B and C),with respectively dripping 100,250,and 500 mM MnCl_(2) solution on the surface of the subscalp skull to explore the best concentration of Mn^(2+) solution in manganese-enhanced magnetic resonance imaging(ME-MRI).Another 12 rats were selected as calcium group to study the possible effect of ionic strength on transport efficiency by adding CaCl_(2),into MnCl_(2).solution.The rats were randomly divided into two groups(group D was performed with 400 mM CaCl_(2),+100 mM MnCl_(2),and group E with 250 mM CaCl_(2),+250 mM MnCl_(2).and MRI examination was performed at 3 h and 24 h after administration.ResultsThe effectiveness of transcranial administration of Mn^(2+)depends on its concentration and drip position,and the penetration of Mn^(2+) was better when the position closed to the cranial suture.The 500 mM concentration of MnCl_(2),produced a larger contrast-to-noise ratio(CNR)(P<0.001),and the hippocampus and thalamus could be clearly displayed 24 hours later.On the contrary,100 mM concentration solution could not cross the skull.It was found that lowconcentration(100 mM)of MnCl_(2)solution could pass through the skull after the addition of Ca^(2+),and the transport efficiency was proportional to the corresponding Ca^(2+)concentration(P<0.001).After transcra-nial application of 500 mM concentration MnCl_(2)at 24 h,ME-MRI showed the best results for neural tissue display.ConclusionMn+solution can enter the brain tissue through the intactskull,and the addition of Ca^(2+)can effectively improve the pen-etration ability of Mn^(2+).The subscalp pathway has a better dis-play effect on nerve tissue,which can provide a relatively less invasive method for ME-MRI of neurve tissue in animal models,expanding the application of Mn^(2+)in neurography.
