摘 要:花生是我国重要的油料和经济作物,非生物胁迫严重影响其产量和品质。乙醇酸氧化酶(Glycolate Oxidase,GOX)不仅是光呼吸途径中的关键酶,同时在植物响应非生物胁迫中发挥重要作用。本研究利用生物信息学的方法,在栽培花生A.hypogaea(2n=40,AABB)全基因组水平上鉴定到了16个GOXs基因家族成员,在野生二倍体花生A.duranensis(2n=20,AA)和A.ipaensis(2n=20,BB)全基因组水平上分别鉴定到了9个和5个GOXs基因家族成员。基因结构分析发现,该基因家族成员由4~20个外显子构成。通过进化树分析,可将花生GOXs基因家族成员分为5个组。以耐盐品种花育25号和盐敏感品种花育20号为材料,进行干旱、盐、高温和低温非生物胁迫处理,根据qRT-PCR分析结果表明,在两个品种和不同组织之间的表达量具有显著差异。本研究为进一步研究花生响应非生物胁迫的机制提供了候选基因。Peanut is one of the economically important oil crops in China.Abiotic stresses affect the yield and quality of peanut crop.Glycolate Oxidases(GOXs)are one of the most important classes of enzymes that are involved in the photorespiration pathway and plays key roles during abiotic stress responses in plant.In this study,16 genes of the GOXs family were identified in cultivated peanut A.hypogaea(2n=40,AABB)genome based on bioinformatics method.Besides,9 and 5 genes were also identified from two wild diploid peanut genomes,A.duranensis(2n=20,AA)and A.ipaensis(2n=20,BB)respectively.Gene structure analysis showed that the GOXs genes are composed of 4 to 20 exons.The GOXs genes were grouped into 5 clusters by phylogenetic tree analysis.Additionally,Huayu20(HY20),salt-susceptible variety,and Huayu25(HY25),salt-tolerant variety,were selected as research materials.The quantitative real-time PCR revealed that GOXs genes from different groups exhibited expression variation in different tissues under drought,salt,heat,and cold stress treatments.These results can provide candidate genes for further functional characterization of GOXs genes responding to abiotic stress in peanut crop.
