摘  要:随着微观操作技术的发展,各种技术被用于细胞变形性的测量,但对非接触和连续测量方法的需求仍然很大。结合脉冲激光光镊和微流控芯片技术,利用流体粘滞阻力使红细胞在捕获时产生一定的形变量,对变形前后的细胞进行图像处理并提取特征参数进行统计分析,测得红细胞的弹性常数为17.95μN/m。同时利用该方法测得流量4μL/h、功率0.3 W下正常红细胞和500μmol/L CaCl溶液处理后红细胞的变形性,统计分析得到其水流方向上的平均形变指数分别约7.05%、10.89%,表明该方法可实现对红细胞变形性的有效表征。该方法为流动条件下测量细胞变形性提供了研究思路。With the development of micro-manipulation techniques,various techniques are used to measure the deformation of cells,but there is still a great demand for non-contact and continuous measurement methods.In this paper,combined with pulsed laser optical tweezers and microfluidic chip technology,the elastic constant of red blood cells was measured to be 17.95μN·mby using fluid viscous resistance to generate certain shape variables during capture,and the image processing of cells before and after deformation and the extraction of characteristic parameters were extracted for statistical analysis,and the elastic constant of red blood cells was measured to be 17.95μN·m.Meanwhile,the deformability of red blood cell treated with 4μl/h flow rate,0.3 w power and 500μmol/L CaClsolution was measured by this method.The average deformability indices in the flow direction were 7.05%and 10.89%,respectively,indicating that this method can effectively characterize the deformability of red blood cells.This method provides a research idea for measuring cell deformability under flow condition.
