摘  要:目的探索促肾上腺皮质激素释放激素(CRH)促进慢性间歇性低氧(CIH)模型小鼠血压升高。方法将雄性C57小鼠随机分为正常氧对照组(Sham)、慢性间歇性低氧组(CIH),慢性间歇性低氧组(CIH)小鼠放入低氧仓进行间歇性低氧处理(每天8 h,连续4周)。HE染色观察脑部病理形态学改变,用Western blot测定PVN中CRH水平,用无创套法测小鼠的尾动脉收缩压(SBP)。结果与对照组(Sham)相比,慢性间歇性低氧4周之后,CIH组小鼠脑部神经元出现细胞肿胀,胞浆空泡化;CRH表达水平升高,SBP明显升高。结论慢性间歇性低氧时,下丘脑室旁核CRH神经元被激活,激活的CRH神经元可增强外周交感神经活动,促进血压升高。Objective To explore the effect of corticotropin releasing hormone(CRH)on increasing blood pressure in mice with chronic intermittent hypoxia(CIH).Method Male C57 mice were randomly divided into normal oxygen control group(Sham),chronic intermittent hypoxia group(CIH),and chronic intermittent hypoxia group(CIH)mice were placed in a hypoxic chamber for intermittent hypoxia treatment(8 hours a day for 4 consecutive weeks).The pathological changes of the brain were observed by HE staining,the CRH level in PVN was measured by Western blot,and the tail artery systolic blood pressure(SBP)of mice was measured by the non-invasive cuff-tail method.Result Compared with the control group(Sham),after 4 weeks of chronic intermittent hypoxia,the brain neurons of the CIH group showed cell swelling and cytoplasmic vacuolation;the expression of CRH increased,and the SBP increased significantly.Conclusion In chronic intermittent hypoxia,CRH neurons in the paraventricular nucleus of the hypothalamus were activated.The activated CRH neurons could enhance peripheral sympathetic nerve activity and promote blood pressure.
