摘  要:目的观察水飞蓟宾对Fas/FasL信号通路的影响,揭示水飞蓟宾促肝星状细胞凋亡的机制。方法将体外培养的肝星状细胞分为正常组、模型组和治疗组,模型组细胞给予PDGF刺激生长,治疗组细胞给予PDGF刺激后加入水飞蓟宾治疗,采用流式细胞术检测肝星状细胞的凋亡,ELISA法检测Fas、Caspase-8蛋白含量。结果模型组肝星状细胞较正常组显著增殖,而治疗组肝星状细胞的凋亡率较模型组明显增加。治疗组Fas、Caspase-8蛋白表达量较模型组明显增加。结论水飞蓟宾可以促进肝星状细胞凋亡,其机制可能与调控Fas/FasL信号通路有关。Objective To discuss the role of silibinin on the apotosis of hepatic stellate cells through observation of the effect of silibinin on Fas/FasL siganal pathway.Method Hepatic stellate cells were cultured in vitro and divided into normal group,model group and treatment group.Model group cells were stimulated with PDGF,treatment group cells were stimulated with PDGF and then treated with silibinin.Flow cytometry was used to detect the apoptosis of HSC,ELISA was used to analyse Fas,Caspase-8 expression.Result The model group HSC growed significantly compared to the normal group.The apoptosis rete of the treatment group was much higher than the model group.The expression of Fas,Caspase-8 protein were much higher than the model group.Conclusion Silibinin could induce HSC apoptosis,the mechanism might be through Fas/FasL signal pathway.
