摘 要:为研究牦牛SRSF10(Serine/argnine-rich splicing factor 10,SRSF10)基因的结构及功能,以美仁牦牛脂肪组织cDNA为模板,克隆牦牛SRSF10基因的编码区(Coding sequence,CDS)序列,并利用基因序列进行生物信息学分析及蛋白理化性质预测。结果表明,美仁牦牛SRSF10基因的CDS区全长789 bp,共编码262个氨基酸;同源性比对发现,牦牛与野牦牛、黄牛之间的SRSF10基因亲缘关系最近(100%),与北极狐最远(97%);牦牛SRSF10蛋白分析显示,SRSF10蛋白内存在2个糖基化位点,1个RRM结构域(74 aa),无跨膜结构域及信号肽区域,其氨基酸等电点11.26,不稳定系数(Ⅱ)101.36,GRAVY=-1.762,预测为稳定的亲水性分泌蛋白;亚细胞定位与核定位预测分析均显示,SRSF10位于细胞核(61.9%),蛋白内共有潜在磷酸化位点63个;SRSF10蛋白高级结构由无规则卷曲(58.02%)、α-螺旋(21.76%)、延伸链(10.69%)和β-转角(9.54%)相互连接而成,主要与同为丝氨酸和富含精氨酸剪接因子、RNA结合蛋白等相关蛋白相互作用。综上所述,SRSF10基因在牦牛脂肪组织中发挥重要作用,该研究结果可为SRSF10基因在牦牛脂肪沉积调控机制的研究提供理论依据,也可为牦牛的遗传育种提供参考。To study the structure and function of the serine/argnine-rich splicing factor 10(SRSF10)gene in yaks,the complete SRSF10 gene CDS sequence was cloned by using the Meiren yak adipose tissue cDNA as the template.The SRSF10 gene nucleotide sequence and physical and chemical properties of protein were analyzed by biological software.The results showed that the CDS region of Meiren yak SRSF10 gene was 789 bp in length and encoded 262 amino acids,the most closely genetic relationship of Meiren yak was the wild yak and Yellow cattle(100%),and the most distant was the arctic fox(97%).The protein analysis of SRSF10 showed that SRSF10 contains 2 glycosylation site,1 RRM domain of 74 aa,no transmembrane structural and no signal peptide region.The amino acid isoelectric point of SRSF10 was 11.26,the instability coefficient(II)was 101.36,and the GRAVY was-1.762.SRSF10 was predicted to be a stably hydrophilic secretory protein.The SRSF10 protein was located in the nucleus(61.9%)by subcellular localization and prediction analysis of nuclear positioning,and contained 63 potential phosphorylation sites.The SRSF10 protein advanced structure consisted of random coil(58.02%),α-helix(21.76%),extended chain(10.69%),andβ-turn(9.54%).In addition,SRSF10 mainly interacted with Serine/arginine-splicing factor protein.In conclusion,SRSF10 gene plays an important role in yak adipose tissue,and this study provides a theoretical basis for the study of SRSF10 gene regulation mechanism of fat deposition in yaks,and can be used as a reference for yaks breeding.
