摘  要:有报道显示,CCNB2与ALDH1L2基因与动物繁殖存在一定的联系,但关于他们在山羊繁殖中的作用还未清晰,为了改进山羊育种,提供有用的遗传标记,首先对3种山羊品种(济宁青山羊,辽宁绒山羊,云上黑山羊)768只母羊CCNB2基因和ALDH1L2基因的8个候选位点进行基因分型,之后再进行群体遗传和关联分析。结果表明,8个候选位点均存在多态性,CCNB2基因中除g.51957276C>T位点在济宁青山羊和辽宁绒山羊中为低度多态性(PIC<0.25)外,其余位点均处于中度多态性(0.25<PIC<0.5);ALDH1L2基因除g.67261430C>G位点在云上黑山羊和济宁青山羊群体中为中度多态性(0.25<PIC<0.5)外,其余位点均为低度多态性(PIC<0.25)。卡方检验结果表明,ALDH1L2基因大部分位点在3个群体中处于Hardy-Weinberg平衡状态(P>0.05),CCNB2基因g.51957276T>C位点在云上黑山羊和济宁青山羊群体中处于Hardy-Weinberg平衡状态(P>0.05),其余位点均处于Hardy-Weinberg不平衡状态(P<0.05)。在云上黑山羊中ALDH1L2基因g.67260707A>T位点TT型个体的产羔数显著高于AA型个体,在窝重上则没有显著性。综上所述,ALDH1L2基因的g.67260707A>T位点的突变可作为云上黑山羊产羔数选择的潜在分子标记。It has been reported that CCNB2 and ALDH1L2 genes are related to animal reproduction,but their role in goat reproduction is not clear.In order to improve goat breeding and provide useful genetic markers,this study first genotyped 8 candidate loci of CCNB2 and ALDH1L2 genes in 768 ewes of three goat breeds(Jining Grey goat,Liaoning Cashmere goat,Yunshang Black goat),and then carried out population genetic and association analysis.The results showed that there were polymorphisms in all 8 candidate loci.In CCNB2 gene,except for g.51957276C>T locus,which was low polymorphism(PIC<0.25)in Jining Grey goat and Liaoning Cashmere goat,the rest were moderate polymorphism(0.25<PIC<0.5);In ALDH1L2 gene,except for the g.67261430C>G locus was moderately polymorphic(0.25<PIC<0.5)in Yunshang Black goat and Jining Grey goat population,the rest were low polymorphism(PIC<0.25).Chi-square test showed that most loci of ALDH1L2 gene were in Hardy-Weinberg equilibrium(P>0.05)in the three populations;However,for the locus of CCNB2 gene,except g.51957276T>C was Hardy-Weinberg equilibrium(P>0.05)in the Yunshang Black goat and Jining Grey goat populations,the rest were in Hardy-Weinberg disequilibrium(P<0.05).In Yunshang Black goats,the litter size of individuals with TT type of ALDH1L2 gene g.67260707A>T was significantly higher than that of AA type,but there was no significant difference in litter weight.To sum up,the mutation of g.67260707A>T in ALDH1L2 gene can be used as a potential molecular marker for litter size selection in Yunshang Black goats.
