摘 要:棘洪滩水库是引黄济青工程唯一的调蓄水库,也是青岛市最重要的饮用水水源地。采用高通量测序方法分析棘洪滩水库春季细菌群落结构多样性、细菌群落结构组成与环境因子的相关性,考察了理化因子对细菌多样性的影响。结果表明,从3月到5月,α多样性分析显示,细菌群落丰富度表现为4月进水口最高,3月出水口最低;细菌群落多样性表现为5月出水口最高,3月进水口最低。β多样性分析显示,3月进水口与出水口细菌群落结构相似性最高,4月进水口和出水口细菌群落结构相似性最低。门水平主要优势菌群为放线菌门(Actinobacteria,34.66%(相对丰度,下同)~57.22%)、变形菌门(Proteobacteria,20.38%~27.08%)、蓝细菌门(Cyanobacteria,0.64%~17.58%)和拟杆菌门(Bacteroides,7.36%~18.56%);属水平的优势菌群为hgcI_clade(14.88%~34.52%)、CL500-29_marine_group(3.13%~15.19%)、绿弯菌属(Chloroflexus,0.16%~13.68%)和孢鱼菌科未知属(g_uncultured_f_Sporichthyaceae,1.27%~16.15%);冗余分析结果表明溶解氧(DO)和化学需氧量(COD)是影响春季棘洪滩水库细菌群落结构的主要环境因子。Jihongtan Reservoir is the only storage reservoir of the Yellow River-to-Qingdao Diversion Project and it is also the most important source of drinking water in Qingdao.High-throughput sequencing was used to analyze bacterial community structural diversity,the correlation between bacterial community structure composition and environmental factors in Jihongtan Reservoir in spring.And the influence of physical and chemical factors on bacterial diversity was discussed.Alpha diversity analysis showed that the bacterial community richness was highest at the water inlet in April and lowest at the water outlet in March;the diversity of the bacterial community was the highest at the water outlet in May and the lowest at the water inlet in March from March to May.Beta diversity analysis showed that the similarity of the bacterial community structure between the water inlet and the water outlet was the highest in March,and the lowest in April.The predominant phylum of bacteria were Actinobacteria(34.66%(relative abundance,the same below)-57.22%),Proteobacteria(20.38%-27.08%),Cyanobacteria(0.64%-17.58%)and Bacteroides(7.36%-18.56%).The predominant genus of bacteria were hgcI_clade(14.88%-34.52%),CL500-29_marine_group(3.13%-15.19%),Chloroflexus(0.16%-13.68%)and g_uncultured_f_Sporichthyaceae(1.27%-16.15%).The results of redundancy analysis showed that DO and COD were the main environmental factors affecting bacterial community structure in spring.
