摘 要:为探索影响葡萄体细胞胚发生的各种条件,以具有强再生能力的‘41B’葡萄(Vitis vinifera Chasselas×Vitis berlandieri)胚性细胞为材料,研究了不同物质对葡萄体细胞胚诱导的影响。结果发现,萘氧乙酸(NOA)和2,4-D两种生长素调节剂对细胞维持和后续体细胞胚诱导效率具有差异;生长素的去除是诱导体细胞胚发生的关键,而活性炭对去除外源生长素的效果非常明显,同时其非选择性吸附对化合物的有效性也存在重要作用。三种细胞分裂素(ZR、TDZ1、6-BA)在体细胞胚诱导过程中促进了非胚性愈伤组织的形成;高浓度丁酸钠(NaB)对胚性细胞具有杀伤作用;透析膜(DM)、高浓度蔗糖和脱落酸(ABA)对体细胞胚诱导及其萌发具有影响。其中,透析膜的使用会导致细胞脱水,高浓度蔗糖和ABA影响体细胞胚出现的同步性。上述结果可为葡萄体细胞胚诱导相关研究中试验处理的设置提供依据。To explore the various conditions affecting the somatic embryogenesis of grapevine,somatic cells of'41B'(Vitis vinifera Chasselas×Vitis berlandieri)with strong regenerative ability was used as materials to study the effects of different substances on grape somatic embryo induction.The results showed that two types of auxins,NOA and 2,4-D had different efficiency in embryonic cells maintenance and subsequent somatic embryo induction;the removal of auxin was the key to inducing somatic embryogenesis successfully,and activated carbon had a very obvious effect on the removal of exogenous auxin,and tis the non-selective adsorption had an important effect on the availability of compounds.Three cytokinins(ZR,TDZ1,6-BA)promoted non-embryonic callus formation during somatic embryo induction;high concentrations of sodium butyrate could cause cell death;dialysis membrane,high concentration of sucrose and ABA affected somatic embryo induction and germination.Among them,the use of dialysis membrane would lead to excessive dehydration of cells;high concentration of sucrose and ABA affected the synchrony of somatic embryogenesis.These results could provide a basis for experimental treatment in the study of grape somatic embryo induction.
