辅酶Q10对NEFA致肝细胞氧化应激损伤的保护作用

摘  要:为了探讨辅酶Q10(CoQ10)对非酯化脂肪酸(NEFA)诱导肝细胞氧化应激损伤的作用,本试验建立了NEFA诱导小鼠肝细胞系AML12氧化应激损伤模型,采用MTT法检测细胞毒性、细胞增殖情况;使用丙二醛(MDA)、总超氧化物歧化酶(T-SOD)、活性氧(ROS)和线粒体膜电位试剂盒检测细胞氧化应激相关指标和线粒体膜电位变化。结果显示,与对照组相比,1.2 mmol/L NEFA(NEFA组)AML12细胞活力极显著降低(P<0.01),而与NEFA组相比,CoQ10+NEFA组AML12细胞活力显著提升(P<0.05);与对照组相比,NEFA组极显著上调AML12细胞MDA和ROS水平,极显著降低AML12细胞T-SOD活性(P<0.01),而与NEFA组相比,CoQ10+NEFA组中细胞MDA含量和ROS水平显著降低、T-SOD活性显著上升(P<0.05);与对照组相比,NEFA组线粒体膜电位极显著下降(P<0.01),而与NEFA组相比,CoQ10+NEFA组细胞线粒体膜电位显著上升(P<0.05)。结果表明,CoQ10可以减轻NEFA引起的小鼠肝细胞氧化应激损伤。In order to evaluate the protective effect of CoQ10 on NEFA-induced oxidative stress injury,this study established the NEFA-induced oxidative stress damage model utilizing mouse hepatocytes AML12.MTT method was used to assess cytotoxicity and cell proliferation;while malondialdehyde(MDA),total superoxide dismutase activity(T-SOD),reactive oxygen species(ROS)and mitochondrial membrane potential kits were used to detect cellular oxidative stress related indicators and mitochondrial membrane potential changes.The results showed that compared to the control group,1.2 mmol/L NEFA(NEFA group)significantly reduced the viability of AML12 cells(P<0.01);while compared to the NEFA group,CoQ10+NEFA significantly increased the viability of AML12 cells(P<0.05);compared to the control group,NEFA significantly upregulated the MDA and ROS levels,but significantly reduced the T-SOD activity in AML12 cells(P<0.01).However,compared to the NEFA group,CoQ10+NEFA significantly reduced the MDA and ROS levels,but significantly increased the T-SOD activity in hepatocytes(P<0.05).Furthermore,compared to the control group,NEFA significantly decreased the mitochondrial membrane potential(P<0.01),whereas compared to the NEFA group,CoQ10+NEFA significantly increased the mitochondrial membrane potential(P<0.05).Thus,CoQ10 can reduce NEFA-induced oxidative stress damage in mice hepatocytes.
