摘 要:Toll样受体(TLRs)是表达于固有免疫细胞的最重要的模式识别受体(PRRs)之一,可通过与不同的病原体相关分子模式(PAMPs)识别而启动固有免疫应答。近年来,在神经炎症性疾病如感染性中枢神经系统疾病、神经退行性疾病、缺血性脑损伤等的研究中,TLRs已经成为人们关注和研究的焦点之一。大量的研究表明,TLRs活化介导的神经炎症反应在脑卒中、阿尔茨海默病(AD)、脑型疟(CM)等神经系统性疾病的发病过程中扮演着非常重要的角色,其参与发病的作用机制十分复杂,因此也造成不同研究对其在神经炎症病程中的作用存在很多争议。而全面地认识TLRs在神经炎症性疾病中的双向调控作用,对相关疾病辅助治疗的药物靶点研究具有重要的推动性意义。本文着重就Toll样受体在几种典型的神经炎症性疾病中的作用与机制进行综述。Toll-like receptors(TLRs)are one of the most important pattern recognition receptors(PPRs)expressed on innate immune cells,which can initiate innate immune responses by recognizing molecular patterns associated with different pathogens(PAMPs).In recent years,TLRs has become one of the focuses of attention and research in the study of neuroinflammatory diseases,such as infectious central nervous system diseases,neurodegenerative diseases,ischemic brain injury and so on.A large number of studies have shown that the neuroinflammatory response mediated by TLRs activation plays a very important role in the pathogenesis of stroke,Alzheimer’s disease(AD),cerebral malaria(CM)and other neurological diseases,and the mechanism involved in the pathogenesis is very complex.Therefore,there are many disputes about the role of TLRs in the course of neuroinflammation in different studies.A comprehensive understanding of the bidirectional regulation of TLRs in neuroinflammatory diseases has important significance for promoting the study of drug targets for adjuvant therapy of related diseases.This paper reviewed the role and mechanism of TLRs in several typical neuroinflammatory diseases.
