摘  要:目的建立超高效液相色谱(UPLC)特征图谱-化学模式识别金银花和山银花的差异性,为金银花和山银花的鉴定提供新的鉴别手段。方法以70%甲醇超声提取金银花和山银花,特征图谱及含量测定利用同一液相色谱条件:乙腈-0.1%磷酸水为流动相,梯度洗脱,同时对新绿原酸、隐绿原酸、绿原酸、木犀草苷、芦丁、金丝桃苷、异绿原酸B、异绿原酸A、异绿原酸C等9种化学成分的色谱峰进行指认和含量测定,并采用SMICA14.1软件进行数据处理、聚类分析和主成分分析。结果建立的金银花UPLC特征图谱中指认了金银花16个共有峰,山银花15个共有峰,聚类分析和主成分分析均可将二者区分开来。二者差异性成分为金丝桃苷、木犀草苷和新绿原酸。结论UPLC特征图谱-化学成分模式识别方法能够全面、有效、直观地反映出金银花和山银花的差异,为金银花及其伪品的鉴别提供新的途径。Objective To distinguish the difference between Jinyinhua(Lonicerae Japonicae Flos)and Shanyinhua(Lonicerae Flos)based on the characteristic chromatogram of ultra performance liquid chromatography(UPLC)-chemical pattern,which provide a new identification method.Methods Jinyinhua(Lonicerae Japonicae Flos)and Shanyinhua(Lonicerae Flos)were extracted with 70%methanol by ultrasonic extraction.The characteristic chromatogram and content determination were carried out by using the same liquid chromatography conditions:acetonitrile-0.1%phosphoric acid water as mobile phase,gradient elution,and simultaneous determination of neochlorogenic acid,cryptochlorogenic acid,chlorogenic acid,luteolin,rutin,hyperoside,isochlorogenic acid B,isochlorogenic acid A and chlorogenic acid C.The chromatographic peaks of 9 chemical components were identified and determined.Smica14.1 software was used for data processing,cluster analysis and principal component analysis.Results 16 common peaks of Jinyinhua(Lonicerae Japonicae Flos)and 15 common peaks of Shanyinhua(Lonicerae Flos)were identified in the UPLC chromatogram,which could be distinguished by cluster analysis and principal component analysis.Hyperoside,luteolin and neochlorogenic acid were the different components.Conclusion UPLC fingerprint pattern recognition method can reflect the differences between Jinyinhua(Lonicerae Japonicae Flos)and Shanyinhua(Lonicerae Flos)comprehensively,effectively and intuitively,and provide a new way for the identification of Jinyinhua(Lonicerae Japonicae Flos)and its counterfeits.
