摘 要:目的对乙型病毒肝炎DNA检测的两种不同核酸提取方法在使用相同的仪器和试剂的条件下进行性能比较,并分析其比较结果.方法仪器采用ABI7500实时荧光核酸扩增仪,试剂采用中山达安公司生产的HBV-DNA检测试剂盒,根据《全国临床检验操作规程》及厂家说明书,分别采用手工法和全自动法提取HBV-DNA进行精密度、准确度、线性范围的性能验证.结果全自动核酸提取法的高值血清的精密度、低值血清的精密度更好(P<0.05);全自动核酸提取法的批间精密度的CV值明显低于手工法(P<0.05).正确度验证结果显示,全自动法的偏倚小于手工法.线性范围验证的结果显示,全自动核酸提取法的相关系数为0.985,优于手工法的相关系数0.959.结论全自动核酸提取法检测HBV-DNA简化了工作流程,优于手工法,能更好地满足实验室自动化检测工作.Ob jective The performance of two different nucleic acid extraction methods for DNA detection of hepatitis B virus was compared using the same instrument and reagent,and the comparison results were analyzed.Methods ABI7500 real-time fluorescent nucleic acid amplitizer was used for the instrument,and HBV-DNA detection kit produced by Zhongshan Daan Company was used for the reagent.According to the"National Clinical Examination Operation Rules"and the manufacturer's instructions,HBV-DNA was extracted by manual method and automatic method respectively to verify the performance of precision,accuracy and linear range.Results The precision of high value serum and low value serum was better by automatic nucleic acid extraction method(P<0.05).CV value of interbatch precision of automatic nucleic acid extraction method was significantly lower than that of manual method(P<0.05).The correctness verification results show that the bias of automatic method is lssthan that of manual method.The results of linear range verification showed that the correlation coeficient of automatic nucleic acid extraction method was 0.985,which was belter than that of manual method.Conc lusion Automatic nucleic acid extraction method for delection of HBV-DNA simplifies the work flow,is better than manual method,can beter meet the laboratory automated detection work.
