摘  要:模拟生物矿化过程以制备骨组织修复材料是目前研究骨组织工程的一种重要途径。文章使用可溶性丝素蛋白作为有机基质,来探究仿生骨组织修复材料制备中的羟基磷灰石的形成过程。在羟基磷灰石的微观形貌表征方面,文章首先进行了扫描电镜观察;其次通过红外光谱法、热重分析法等简单快捷的表征技术,来分析羟基磷灰石的生长过程。结果表明:丝素蛋白对磷酸钙晶体的矿化过程具有显著影响。丝素蛋白通过形核作用控制磷酸钙矿化过程,加速生物矿化过程中透磷酸钙(Dicalcium phosphate dehydrate,DCPD)向羟基磷灰石(Hydroxyapatite,HA)的晶相转变。Bone tissue engineering materials based on calcium phosphate mineralization have been studied widely.The biomimetic strategy provides a platform to explore the natural bone composition and structure.It is demonstrated that the organic matrix-modulated bio mineralization is a classic and hot topic in bone-based biomedical research.It should be noted that the molecular weight,molecular structure,and reactant concentration of organic matter would affect the growth of calcium phosphate crystals during bio mineralization.As a natural material,silk fibroin exhibits excellent human biosafety and is widely used for bone tissue engineering composite.Currently,silk fibroin has been commonly used to prepare calcium phosphate composites.However,its function in calcium phosphate crystal growth during bio mineralization is still unclear.In this study,silk fibroin was used as an organic matrix regulator to detect the influence of calcium phosphate crystals during bio mineralization.Several features of nano-bydroxy apatite were characterized by Faurier transform infrared spectroscopy(FTIR),thermo gravimetry(TG),and scanning electron microscope(SEM).The results show that silk fibroin plays a significant role in calcium phosphate bio mineralization.Silk fibroin,as the organic matrix regulator,shows the potential property to regulate calcium phosphate bio mineralization.During the bio mineralization,the interactions between calcium ions and silk fibroin initially influence the nucleation of calcium phosphate,which will further accelerate the crystal transition of dicalcium phosphate dehydrate(DCPD) to hydroxyapatite(HA).In addition,the silk fibroin composites can significantly affect the bio mineralization of calcium phosphate,inducing the micro-scale HA/silk fibroin composites.The crystalline phase of HA deposited on silk fibroids has a preferred orientation in the(002) C axis direction within the composites.It is demonstrated that hydroxyl,carbonyl,and other groups in the organic matrix affect the bio mineralization and nucleatio
