摘 要:为探究太子参种质资源的遗传多样性及亲缘关系,采用简单重复序列区间多态性(ISSR)和随机扩增多态性(RAPD)标记对来自不同产地的10个太子参种质进行遗传多样性分析。结果表明,13条ISSR引物和16条RAPD引物在10个太子参种质中分别获得101、89个扩增位点。其中多态性位点数(NPL)分别为58、63个,多态性位点频率(PPL)分别为57.37%、70.32%。基于ISSR标记的太子参种质间的观测等位基因数(Na)、有效等位基因数(Ne)、Nei’s遗传多样性指数(H)和Shannon’s多态性指数(I)的平均值分别为1.5737、1.3443、0.3532、0.5297。基于RAPD标记的太子参种质间的Na、Ne、H和I的平均值分别为1.7032、1.4181、0.3549、0.5321。综合2种标记的GS为0.668~0.911。综合2种标记的聚类结果表明,当GS为0.780时,10个太子参种质可聚为4组,聚类分析结果与太子参种质的地理分布有明显的相关性。In order to explore the genetic diversity and genetic relationship of Pseudostellaria heterophylla germplasm resources,inter-simple sequence repeat(ISSR)and randomly amplified polymorphic DNA(RAPD)markers were used to analyze the genetic diversity of 10 Pseudostellaria heterophylla germplasms from different habitats.The results showed that 13 ISSR primers and 16 RAPD primers obtained 101 and 89 amplification sites in 10 germplasms of Pseudostellaria heterophylla.The number of polymorphism loci(NPL)were 58 and 63,and the percentage of polymorphism loci(PPL)were 57.37%and 70.32%respectively.The average values of observed number of alleles(Na),effective number of alleles(Ne),Nei’s genetic diversity index(H)and Shannon’s information index(I)among Pseudostellaria heterophylla germplasms based on ISSR markers were 1.5737,1.3443,0.3532 and0.5297,respectively.The average values of Na,Ne,H and I among Pseudostellaria heterophylla germplasms based on RAPD markers were 1.7032,1.4181,0.3549 and 0.5321,respectively.The GS between Pseudostellaria heterophylla germplasms ranged from 0.668 to 0.911 based on the two markers.The clustering results based on the synthesis of the two markers showed that 10 germplasms of Pseudostellaria heterophylla could be clustered into 4 groups when GS was 0.780,and the clustering results were significantly correlated with the geographical distribution of Pseudostellaria heterophylla germplasms.
