摘  要:目的探讨肝细胞癌中铁死亡的作用机制及其相关长链非编码RNA(lncRNA)的预后价值。方法从TCGA数据库下载肝癌转录组数据,筛选铁死亡差异基因并进行富集分析;相关性检验获取与铁死亡相关的lncRNA;使用Cox回归分析构建预后模型。根据风险评分公式计算风险得分,并将队列中的肝癌患者分为高、低风险组,并对其进行生存分析及免疫差异分析。结果筛选出84个铁死亡差异基因。富集分析显示与mTOR代谢、糖酵解、γ干扰素应答等相关,纳入17个铁死亡lncRNA构建预后模型。低风险组预后优于高风险组(P<0.01);受试者操作特征曲线表明该模型具有良好的预测能力。免疫微环境分析发现,多种免疫细胞具有浸润差异,且鉴定出17个具有表达差异的免疫检查点。结论铁死亡基因及相关lncRNA在肝癌中都起着重要的调控作用,且免疫微环境的变化也是肝癌治疗的潜在靶标。Objective To investigate the mechanism of ferroptosis in hepatocellular carcinoma and the prognostic value of long non-coding RNA(lncRNA).Methods Hepatocellular carcinoma transcriptome data were downloaded from TCGA database,ferroptosis differential genes were screened and enriched analyzed;lncRNA associated with ferroptosis was obtained by correlation test;Cox regression analysis was used to construct a prognostic model.According to the risk scoring formula,the risk scores were calculated,and the patients with liver cancer were divided into high,low risk groups,and survival analysis and immune differential analysis were performed.Results Eighty-four ferroptosis differential genes were selected.Enrichment analysis showed correlation with mTOR metabolism,glycolysis,and γ interferon respond,and 17 ferroptosis lncRNAs were included to construct a prognostic model.The prognosis of low-risk group was better than that of high-risk group(P<0.01);receiver operating characteristic curve showed that the model had good predictive ability.The analysis of immune microenvironment revealed that a variety of immune cells had different infiltration,and 17 immune checkpoints with different expression were identified.Conclusion Both ferroptosis genes and related lncRNAs play important regulatory roles in liver cancer,and the changes of immune microenvironment are also potential targets for liver cancer treatment.
