摘 要:目的:采用多指标-Box-behnken design(BBD)效应面法,优选赤芍-川芎药对(1∶1)最佳提取工艺。方法:在单因素试验的基础上,采用三因素三水平BBD试验设计对赤芍-川芎(1∶1)中4成分成分提取工艺参数进行优选。以乙醇浓度、料液比、提取时间为自变量,以芍药内酯苷、芍药苷、洋川芎内酯A及藁本内酯含量为因变量,采用Hassan方法计算总评“归一值”,并用多元线性回归及二项式拟合,建立总评归一值与自变量之间的数学关系,经效应面法预测最佳工艺条件。结果:最佳提取工艺为乙醇浓度68%、料液比1∶11、提取时间79 min,提取2次;芍药内酯苷、芍药苷、洋川芎内酯A及藁本内酯含量分别为7.5870,46.1920,8.7840,25.7310 mg·g^(-1)。结论:实测值与预测值接近,表明采用多指标-效应面法优化后得到的综合提取工艺参数可用于赤芍-川芎药对(1∶1)中有效成分的提取。Objective:To optimize the extraction process of drug pair Paeoniae Radix Rubra-Ligusticum chuanxiong by multiple indicators-response surface method.Methods:On the basis of single factor screening,a three-factor and three-level Box-Behnken experimental design was employed with ethanol concentration,liquid ratio and extraction time as independent variables.Dependent variables were transformed into desirabilities mathematically by Hassan’s method.Data of overall desirabilities were fitted to a second order polynomial equation,through which three dimensional response surface graphs were obtained.Optimum experimental conditions were selected from the stationary point of the response surfaces.Results:Optimum process conditions were as follows:11-fold amount of 68%ethanol,extracted twice with 79 min for each time.The contents of albiflorin,paeoniflorin,senkyunolide A and ligustilide were 7.5870,46.1920,8.7840 and 25.7310 mg·g^(-1),respectively.Conclusion:The measured value is close to the predicted value,which indicates the comprehensive extraction parameters optimized by multiple indicators-response surface method can be used for the determination of albiflorin,paeoniflorin,senkyunolide A and ligustilide in Paeoniae Radix Rubra-Ligusticum chuanxiong.
