携CX3CL1抗体的靶向超声造影剂在卵巢癌腹腔转移中的实验研究

摘　　要：目的 探讨携CX3CL1抗体的靶向超声造影剂在卵巢癌腹腔转移中的诊断价值。方法 6~8周裸鼠50只随机取10只设为空白对照组。剩余40只建立卵巢癌腹腔转移模型,建模成功后分为4组实验组,每组各10只。均接受非靶向造影剂瘤体成像,间隔30 min后采用携带CX3CL1抗体的靶向微泡造影剂瘤体成像,分析各时间强度曲线(TIC)的峰值强度(PI)和达峰时间(TTP);采用Pearson相关性分析软件对CX3CL1表达量与靶向超声造影参数进行相关性分析。结果 实验组携CX3CL1抗体的靶向超声造影剂PI高于非靶向造影剂瘤体成像(P<0.05);非靶向造影剂瘤体成像与携CX3CL1抗体的靶向超声造影剂TTP差异具有统计意义(P<0.05),非靶向造影剂瘤体成像下TTP高于携CX3CL1抗体的靶向超声造影剂;相关性分析结果表明:裸鼠卵巢癌腹腔移植瘤中CX3CL1表达量与靶向超声造影剂中TTP呈负相关性(P<0.05)。结论 携CX3CL1抗体的靶向超声造影剂在卵巢癌腹腔转移中的诊断价值优于非靶向造影剂。Objective To investigate the diagnostic value of targeted ultrasound contrast agent carrying CX3 CL1 antibody in ovarian cancer abdominal metastasis. Methods Ten of 50 nude mice at 6-8 weeks age were randomly selected as a blank control group. The remaining 40 nude mice were divided into four experimental groups with ten nude mice after established as ovarian cancer abdominal metastasis models. All mice received non-targeted contrast tumor imaging, and targeted microbubble contrast tumor imaging carrying CX3 CL1 antibody was performed after a 30 min interval, then the peak intensity(PI) and time to peak(TTP) of each time intensity curve(TIC) were analyzed. Pearson correlation was used to analysis the correlation between CX3 CL1 and targeted ultrasonography parameters. Results Targeted ultrasound contrast PI with CX3 CL1 antibody was higher in the experimental group than that of non-targeted contrast tumor imaging(P<0.05). There was statistically significant difference between the TTP of between non-targeted contrast tumor imaging and targeted ultrasound contrast with CX3 CL1 antibodies(P<0.05), and the TTP of non-targeted contrast tumor imaging was higher than that in targeted ultrasound contrast with CX3 CL1 antibodies(P<0.05). The correlation analysis showed that CX3 CL1 expression in ovarian cancer with peritoneal metastasis in nude mice was negatively correlated with the TTP in targeted ultrasound contrast imaging(P<0.05). Conclusions The diagnostic value of targeted ultrasound contrast agent with CX3 CL1 antibody is better than that of non-targeted contrast agent in ovarian cancer with abdominal metastasis.
