摘 要:目的探讨1,25-二羟基维生素D_(3)[1,25(OH)_(2)D_(3)]对DM小鼠周围神经功能的影响。方法C57BL/6J小鼠腹腔注射STZ构建DM小鼠模型,随机分为DM组(DM)、DM第1周1,25(OH)_(2)D_(3)治疗组(1w-DM)、DM第2周1,25(OH)_(2)D_(3)治疗组(2w-DM)、DM周围神经病变1,25(OH)_(2)D_(3)治疗组(DPN)以及DM第1周溶剂对照组(1w-DMC)、DM第2周溶剂对照组(2w-DMC)、DPN溶剂对照组(DPNC),每组各15只,并设置正常对照组(NC)10只。建模后第1、2、4周分别对1w-DM、2w-DM、DPN组予1,25(OH)_(2)D_(3)腹腔注射,连续5 d;监测各组血糖、体重、触觉、热痛觉及协调能力,8周后收集血清检测25(OH)D_(3)水平。结果与DM组比较,1w-DM、2w-DM组第8周体重升高(P<0.05),血糖降低(P<0.05),协调运动时间、血清25(OH)D_(3)水平升高(P<0.05或P<0.01),触觉反应时间、热痛觉反应时间更短(P<0.05)。结论在DM神经功能受损前,1,25(OH)_(2)D_(3)治疗可有改善DM小鼠周围神经功能。Objective To evaluated the effects of 1,25-dihydroxyvitamin D_(3)[1,25(OH)_(2)D_(3)]on peripheral nerve function in DM mice.Methods C57BL/6J mice were intraperitoneally injected with STZ to establish DM mouse model.They were randomly divided into DM group(DM),1st week 1,25(OH)_(2)D_(3)treatment group(1w-DM),2nd week 1,25(OH)_(2)D_(3)treatment group(2w-DM),diabetic peripheral neuropathy1,25(OH)_(2)D_(3)treatment group(DPN),1st week DM solvent control group(1w-DMC),2nd week DM solvent control group(2w-DMC),DPN solvent control group(DPNC)with 15 in each groups,and 10 in normal control group(NC).1w-DM group,2w-DM group and DPN group were intraperitoneally injected with 1,25(OH)_(2)D_(3)for 5 consecutive days at week 1,2 and 4 after model construction.Blood glucose,body weight,touch,heat pain sensation and coordination ability of mice were monitored in each group for 8 weeks.After 8 weeks,serum was collected for 25(OH)D_(3)testing.Results After 8 weeks,compared with DM group,the body weight increased,blood glucose decreased(P<0.05),coordinated motion time and serum25(OH)D_(3)level increased(P<0.05 or P<0.01),tactile response time and heat pain response time were shorter in 1w-DM and 2w-DM groups(P<0.05).Conclusion 1,25(OH)_(2)D_(3)treatment can improve peripheral nerve function in DM mice before neurological impairment.
