摘 要:为探究延黄牛CD44基因及短支链酰基辅酶A脱氢酶(ACADSB)基因多态性及其与肉质性状的相关性,选取54头延黄牛为研究对象,采用PCR扩增产物Sanger测序技术检测单核苷酸多态性(SNP)位点,利用“牛等家养动物群体遗传分析工具V1.0”评价延黄牛群体遗传结构,利用SPSS 19.0软件分析基因多态性与肉质性状的相关性。结果:延黄牛CD44基因外显子17 g.65736591处存在C/T突变,延黄牛ACADSB基因外显子1 g.42806106处存在A/G突变;卡方检验显示,检测到的SNP位点在延黄牛群体中均处于Hardy-Weinberg平衡状态(P>0.05),且在延黄牛群体中变异较小,均属于中度多态(多态信息含量为0.25~0.5);关联分析结果显示,延黄牛CD44基因SNP位点不同基因型间的肉质性状均无显著差异(P>0.05),ACADSB基因SNP位点不同基因型间在压榨水分、解冻损失率及肉红度(A)值间存在显著性差异(P<0.05,P<0.01),而在其他肉质性状间无显著差异,初步说明CD44和ACADSB基因有望作为延黄牛肉质性状的优良候选基因。In order to identify the polymorphism of CD44 molecules and the Acly-CoA dehydrogenase short/branched chain(ACADSB) and their correlation with meat quality traits in Yanbian Yellow cattle, 54 Yanbian Yellow cattle were selected as research subjects. Single nucleotide polymorphism(SNP) sites were detected by PCR amplification product Sanger sequencing. The genetic structure of Yanbian Yellow cattle was evaluated by“Cattle and other Domestic Animal Population Genetic Analysis Tool V1. 0”,and the correlation between the genetic polymorphism and meat quality traits in the cattle was analyzed by SPSS 19. 0. The results showed that there was C/T mutation in CD44 exon 17 g. 65736591,and A/G mutation in ACADSB exon 1 g. 42806106. The CHI-square test showed that all the SNP loci were in a Hardy-Weinberg equilibrium state in the Yanbian Yellow cattle( P>0. 05),but their variation was small,being in moderate polymorphism( 0. 25-0. 5). The result of association analysis showed that,there were no significant correlation among difference in meat quality among different genotypes of CD44 SNP site( P>0. 05),there were( extremely) significant differences in pressing water,thawing loss rate and meat redness( A) value among different genotypes of ACADSB SNP site( P<0. 05,P<0. 01),but no significant differences in other meat quality traits. The results indicated preliminarily indicated that CD44 and ACADSB genes were expected to be good candidate genes for meat quality traits of Yanbian Yellow cattle.
