摘　　要：目的探讨诃子提取物对不同转移性乳腺癌细胞株增殖、凋亡的影响。方法选取不同转移性的乳腺癌细胞株即MCF-7(低转移性乳腺癌细胞)、MDA-MB-231(中转移性乳腺癌细胞)、MDA-MB-435(高转移性乳腺癌细胞)以及人正常乳腺上皮细胞HBL-100为研究对象,将对数生长期的MCF-7、MDA-MB-435、MDA-MB-231、HBL-100细胞分别进行分组,记为对照组(未处理的细胞)、诃子提取物高浓度组(125μg/ml)、诃子提取物中浓度组(93.75μg/ml诃子提取物)、诃子提取物低浓度组(62.5μg/ml诃子提取物)。流式细胞术检测各组细胞凋亡;Transwell实验检测各组细胞侵袭与迁移;MTT法检测各组细胞增殖情况;Western blot检测各组细胞中基质金属蛋白酶2(MMP-2)、B淋巴细胞瘤-2相关蛋白(Bax)、细胞周期素D1(CyclinD1)蛋白表达情况。结果诃子提取物对HBL-100细胞增殖无显著影响(P>0.05);与对照组相比,诃子提取物低、中、高浓度组乳腺癌细胞的侵袭和迁移数目、细胞增殖率、MMP-2、CyclinD1均显著降低,Bax蛋白表达、细胞凋亡率显著增加(P<0.05)。结论诃子提取物可以诱导不同转移性乳腺癌细胞株凋亡,抑制其增殖、侵袭和迁移。Objective To investigate the impacts of myrobalan extract on the proliferation and apoptosis of different metastatic breast cancer cell lines.Methods Different metastatic breast cancer cell lines were collected as the research objects,including MCF-7(low metastatic breast cancer cells),MDA-MB-231(moderately metastatic breast cancer cells),MDA-MB-435(highly metastatic breast cancer cells)and human normal breast epithelial cells HBL-100,the MCF-7,MDA-MB-435,MDA-MB-231,and HBL-100 cells in the logarithmic growth phase were grouped into the control group(untreated cells),the high concentration group of myrobalan extract(125μg/ml),medium concentration group of myrobalan extract(93.75μg/ml myrobalan extract),and low concentration group of myrobalan extract(62.5μg/ml myrobalan extract).Flow cytometry was performed to detect cell apoptosis in each group;Transwell assay was performed to detect the invasion and migration of cells in each group;MTT method was performed to detect the proliferation of cells in each group;Western blot was performed to detect the protein expressions of matrix metalloproteinase 2(MMP-2),B-lymphoma-2-associated protein(Bax)and cyclin D1(CyclinD1)in the cells of each group.Results Myrobalan extract had no significant effect on the proliferation of HBL-100 cells(P>0.05);compared with the control group,the numbers of invasion and migration,cell proliferation rate,MMP-2 and CyclinD1 of breast cancer cells in low,medium and high concentration groups of myrobalan extract were significantly decreased,the expression of Bax protein and the apoptosis rate were significantly increased(P<0.05).Conclusion Myrobalan extract can induce apoptosis and inhibit the proliferation,invasion and migration of different metastatic breast cancer cell lines.
