摘  要:目的观察电针对急性心肌缺血(AMI)大鼠铁死亡-脂质代谢通路的影响,探讨电针保护心肌细胞的作用机制。方法将SD大鼠随机分为伪手术组、模型组和电针组,每组15只。结扎冠状动脉复制AMI模型,电针组给予电针神门-通里段,每日1次,每次30 min,连续3 d,剩余两组不予治疗。电针结束后采集大鼠腹主动脉血液和心肌组织。用Powerlab记录大鼠心电图;ELISA法检测大鼠血清肌钙蛋白I(cTn-I)含量;HE染色对心肌组织做病理观察;化学荧光法检测心肌组织活性氧(ROS)水平;Western blot检测心肌组织长链脂酰辅酶A合成酶4(ACSL4)、磷脂胆碱酰基转移酶3(LPCAT3)、谷胱甘肽过氧化物酶4(GPX-4)蛋白表达;RT-PCR检测ACSL4、LPCAT3、GPX-4 mRNA的表达;比色法检测心肌组织谷胱甘肽(GSH)水平。结果与伪手术组比,模型组大鼠心电图ST段抬高;血清cTn-I上升(P<0.05);心肌纤维排列紊乱,纤维间隙增宽,细胞肿胀明显;心肌组织中ROS水平升高(P<0.05);ACSL4、LPCAT3蛋白及mRNA表达升高(P<0.05),GPX-4蛋白及mRNA表达降低(P<0.05);GHS水平降低(P<0.05)。与模型组相比,电针组大鼠心电图ST段下降;血清cTn-I降低(P<0.05);心肌纤维间隙缩小,细胞肿胀减轻;心肌组织中ROS水平降低(P<0.05);ACSL4、LPCAT3蛋白及mRNA表达降低(P<0.05),GPX-4蛋白和mRNA表达升高(P<0.05);GHS水平升高(P<0.05)。结论电针可能通过升高细胞内GSH水平,激活GPX-4系统,抑制铁死亡脂质代谢通路中ROS水平以及ACSL4、LPCAT3活性,发挥保护心肌细胞作用。Objective To observe the effect of electroacupuncture on ferroptosis-lipid metabolism pathway in myocardial cells of rats after myocardial ischemia(AMI),and to explore the positive effect of acupuncture on AMI.Methods According to the principle of random grouping,SD rats were divided into sham operation group,model group and electroacupuncture group,each group of 15 cases only.Coronary artery ligation method to copy the AMI model,electroacupuncture group was given cupping treatment Shenmen(HT7)-Tongli(HT5),once a day,every time 30 min,continuous 3 day,the rest of the two groups of untreated.Blood and myocardial tissue of abdominal aorta of rats were collected after electroacupuncture.ECG of rats were recorded by Powerlab.Serum troponin I(cTn-I)was detected by ELISA.HE staining was used for pathological observation of myocardial tissue.Chemical fluorescence detection of myocardial tissue ROS levels.WB was used to detect the expression of ACSL4,LPCAT3,GPX-4 in myocardium.The expression of ACSL4,LPCAT3 and GPX-4 mRNA were detected by PCR.Colorimetry to detect myocardial tissue GSH levels.Results Compared with sham operation,ST segment elevation of ECG,serum cTn-I increased in model group(P<0.05);The arrangement of myocardial fibers was disordered,the fiber space was widened,and the cell swelling was obvious;ROS level in myocardial tissue increased(P<0.05);The expression of ACSl4 and LPCAT3 protein and mRNA increased(P<0.05),while the expression of GPX-4 protein and mRNA decreased(P<0.05);The level of GHS decreased(P<0.05).Compared with the model group,the ST segment of ECG in the electroacupuncture group decreased;Serum cTn-I decreased(P<0.05);The gap between myocardial fibers was narrowed and the swelling of cells was reduced;The level of ROS in myocardial tissue decreased(P<0.05);The expression of ACSl4 and LPCAT3 protein and mRNA decreased(P<0.05),while the expression of GPX-4 protein and mRNA increased(P<0.05);The level of GHS increased(P<0.05).Conclusion Electroacupuncture may increase intracellular GSH levels,
